超氧化物歧化酶1基因多态性与2型糖尿病患者周围血管病变风险的相关性 被引量:9

Genetic polymorphisms of superoxide dismutase 1 are associated with risk of peripheral vascular disease in patients with type Ⅱ diabetes mellitus

摘要 目的研究超氧化物歧化酶1(superoxide dismutase 1,SOD1)基因多态性与2型糖尿病(type 2 diabetes mellitus,T2DM)发生周围血管病变风险的相关性。方法选取2015年7月-2017年7月承德医学院附属医院内分泌科收治的360例T2DM患者为研究对象,根据是否患有周围血管病变将其分为观察组(有周围血管病变)和对照组(无周围血管病变)。采用Sequenom MassARRAY SNP基因分型技术对SOD1基因上rs2234694、rs17880487和rs74315452 3个SNP进行分型。结果本研究中观察组和对照组各180例,2组间性别、年龄、体质指数(BMI)、T2DM病程、收缩压、舒张压、合并患高血压、吸烟情况的差异均无统计学意义(P>0.05)。Rs2234694、rs17880487和rs74315452 3个SNP均符合Hardy-Weinberg平衡定律(P-HWE>0.05)。观察组中rs2234694、rs17880487和rs74315452的最小等位基因C、T和C的频率均明显高于对照组(P <0.05)。观察组rs2234694位点的AC和CC基因型频率、rs17880487位点的CT和TT基因型频率、rs74315452位点的TC和CC基因型频率均高于对照组(P <0.05)。Rs2234694位点在显性和隐性遗传模型下均与增加T2DM患者周围血管病变风险具有相关性(P <0.05)。Rs17880487位点在显性和隐性模型下均与增加T2DM患者周围血管病变风险具有相关性(P <0.05)。Rs74315452位点在隐性和加性模型下均与增加T2DM患者周围血管病变风险具有相关性(P <0.05)。结论 SOD1基因上rs2234694、rs17880487和rs74315452 3个SNP均与增加T2DM患者发生周围血管病变风险具有相关性。 Objective To investigate the association between genetic polymorphisms of superoxide dismutase1(SOD1)and risk of peripheral vascular disease in patients with type II diabetes mellitus(T2DM).Methods A total of360patients with typeⅡdiabetes mellitus(T2DM)were enrolled in the Affiliated Hospital of Chengde Medical University from July2015to July2017.According to whether there is peripheral vascular disease,the participants were divided into observation group and control group.Three SNPs of rs2234694,rs17880487and rs74315452on SOD1gene were genotyped by Sequenom MassARRAY SNP genotyping technique.Results There were180cases in the observation group and the control group,respectively.There was no significant difference in gender,age,BMI,T2DM duration,systolic blood pressure,diastolic blood pressure,hypertension,and smoking between the two groups(P>0.05).All of three SNPs are in accordance with the Hardy-Weinberg equilibrium law(P-HWE0.05).The frequencies of the minor alleles C,T and C of rs2234694,rs17880487and rs74315452in the observation group were significantly higher than those in the control group(P<0.05).The AC and CC genotype frequencies of the rs2234694,the CT and TT genotype frequencies of the rs17880487,and the TC and CC genotype frequencies of the rs74315452in the observation group were higher than those in the control group(P<0.05).The rs2234694was associated with increased risk of peripheral vascular disease in patients with T2DM in both dominant and recessive genetic models(P<0.05).The rs17880487was associated with increased risk of peripheral vascular disease in patients with T2DM in both dominant and additive models(P<0.05).The Rs74315452was associated with increased risk of peripheral vascular disease in patients with T2DM in both recessive and additive models(P<0.05).Conclusion Rs2234694,rs17880487and rs74315452in SOD1gene were all associated with increased risk of peripheral vascular disease in patients with T2DM.
