慢性心力衰竭病人血清miR-451a调控MMP-9的表达及临床意义 被引量:11

摘要 目的探讨慢性心力衰竭病人血清miR-451a与基质金属蛋白酶-9(MMP-9)的表达情况及其临床意义。方法选取我院确诊为慢性心力衰竭的病人80例作为研究组,美国纽约心脏病学会(NYHA)分级:Ⅰ级20例,Ⅱ级22例,Ⅲ级26例,Ⅳ级12例。另选择健康成年人80名作为对照组。应用实时荧光定量PCR(qRT-PCR)和酶联免疫吸附(ELISA)法分别检测血清miR-451a和MMP-9表达水平,分析二者在不同NYHA分级中的表达情况及相关性。结果与对照组比较,研究组miR-451a表达水平显著降低,而血清MMP-9表达水平显著增加,差异均有统计学意义(P <0.05)。随着NYHA分级的增高,miR-451a表达水平下降,而MMP-9表达水平上升,各分级间差异均有统计学意义(P <0.05)。线性相关分析发现,随着miR-451a表达水平的降低,MMP-9表达水平逐渐增加,血清miR-45a与MMP-9的表达呈负相关(r=-0.702,P <0.05)。结论 miR-451a至少部分通过调控MMP-9表达而在慢性心力衰竭的发生发展及病情演变过程中起作用。 Objective To investigate the expression of serum miR-451a and matrix metalloproteinase-9(MMP9)in patients with chronic heart failure(CHF),and to analyze its clinical significance.Methods Eighty patients with CHF,which included20cases of gradeⅠ,22cases of gradeⅡ,26cases of gradeⅢ,and12cases of gradeⅣby New York Heart Association(NYHA)classification criteria,were selected as the study group.Eighty healthy adults were selected as the control group.The levels of miR-451a and MMP-9were detected by real time quantitative PCR(qRT-PCR)and enzyme linked immunosorbent assay(ELISA).The expression and correlation were analyzed among different NYHA grades.Results Compared with the control group,the expression level of miR-451a in the study group was significantly decreased,while the level of serum MMP-9expression was significantly increased(P<0.05).There were differences in the expression levels of miR-451a and MMP-9among the grades(P<0.05).With the progress of the NYHA classification,the expression of miR-451a decreased,while the expression of MMP-9was increased,and the differences between the NYHA classification were statistically significant(P<0.05).The linear correlation analysis showed that the expression level of MMP-9increased gradually with the decrease of miR-451a expression level,and the expression of serum miR-451a was negatively correlated with MMP-9(r=-0.702,P<0.05).Conclusion The miR-451a plays a role in the development and progression of CHF at least in part by regulating the expression of MMP-9.
