甘蔗脱毒健康种苗中蔗糖转运蛋白基因的差异表达分析 被引量:1

摘要 蔗糖转运蛋白与植物糖分积累和产量形成密切相关,研究甘蔗脱毒健康种苗中的蔗糖转运蛋白基因的表达特性,有助于揭示其增产增糖的分子机制。采用Real-time PCR技术分析比较3个蔗糖转运蛋白家族基因SUT1、SUT4和SUT6在不同生长时期甘蔗脱毒健康种苗和未脱毒种苗中的表达差异。结果表明,SUT1、SUT4和SUT6在甘蔗脱毒健康种苗苗期未成熟叶片中上调表达、在成熟叶片中SUT1和SUT4下调表达,SUT6上调表达;在分蘖期脱毒健康种苗未成熟和成熟叶片中SUT1、SUT4和SUT6均上调表达;在拔节期脱毒健康种苗中SUT1表达差异不明显,而SUT4和SUT6在未成熟叶片和茎节中上调表达;在成熟期脱毒健康种苗未成熟叶片、成熟叶片、未成熟茎节中SUT1、SUT4和SUT6均上调表达。总之,SUT1、SUT4和SUT6在甘蔗脱毒健康种苗快速生长、代谢旺盛的组织中被上调表达。初步推测甘蔗种苗经过脱毒处理,能够在代谢活跃组织中上调蔗糖蛋白基因的表达,促进糖分在甘蔗体内的分配和利用,最终提高产量和糖含量。 Sucrose transporters are closely correlative with sugar storage and crop yield formation.It is helpful to clarify the molecular mechanism on increasing yield and sucrose content by analyzing the expression pattern of sucrose transporter genes in disease free sugarcane seedlings.The expression differences of three sucrose transporter genes,SUT1,SUT4and SUT6were analyzed between sugarcane diseases free seedlings and ordinary seedlings during the whole growth period by Real time PCR.The results showed that SUT1,SUT4and SUT6were up regulated in immature leaves of diseases free sugarcane plants at seedling growth stage,however SUT1and SUT4were down regulated and SUT6were up regulated in mature leaves of diseases free sugarcane plants at seedling growth stage.SUT1,SUT4and SUT6were up regulated in leaves of diseases free sugarcane plants at tillering growth stage.SUT4and SUT6were up regulated in immature leaves and internodes of diseases free sugarcane plants at elongation growth stage.The expression of SUT1was not different between diseases free and ordinary sugarcane plants at elongation growth stage.Moreover,SUT1,SUT4and SUT6were up regulated in leaves and immature internodes of diseases free sugarcane plants at mature stage.In conclusion,SUT1,SUT4and SUT6were up regulated in actively metabolism and rapid growth tissues of diseases free sugarcane plants.It postulated that the up-regulated sucrose transporter genes by disease-free treatment promoted sucrose partition and utilization to ultimately improve the cane yield and sucrose content.
