马立克病病毒感染鸡红细胞6种免疫相关因子转录水平的鉴定 被引量:1

Identification of Transcription Levels of Six Immune-related Factors in Chicken Erythrocytes Infected with Marek's Disease Virus

摘要 为了探究在感染马立克病病毒(MDV)后羽髓中病毒增殖情况以及红细胞中免疫相关因子的免疫应答反应,将4日龄雏鸡腹腔注射MDV超强毒株MD5,在攻毒后不同时间进行羽毛和血样采集,通过实时荧光定量PCR检测羽髓病毒载量和红细胞中6种免疫相关基因的mRNA表达水平。结果显示,在攻毒后第7天检测到MDV拷贝,并且14天检测到的拷贝数显著高于其他检测时间;与第14天对照组相比,攻毒组COX2的表达显著上调(P<0.05),而在第22天COX2的表达整体高于第14天(P<0.05);在第14和22天,PTGER4、HPGDS和GSTA3在攻毒组中的表达量显著低于对照组(P<0.05);与之相反,在第14天和第22天,PTGR1在攻毒组中的表达量显著高于对照组(P<0.05);第14天攻毒组的Ex-FABP表达水平显著低于对照组,而在第22天攻毒组显著高于对照组(P<0.05)。这表明感染MDV后红细胞中6种免疫基因发生显著的免疫应答,并且红细胞可能通过调节相关的免疫反应而在MDV感染过程中发挥一定的免疫调控作用。 To explore the virus proliferation in the feather tips and the immune response of these six immune-related genes in MDV-infected erythrocytes,four-day-old chicks were intra-abdominally infected with MD5at a dose of500PFU.The feathers and blood samples were collected on different sampling time points.The virus load and the mRNA levels of these six immune-related genes in the erythrocytes were detected by real-time PCR.It was shown that the MDV was detected after7d.p.i.and the MDV copy number at14d.p.i.was significantly higher than that of all other sampling time points;The mRNA expression of COX2was significantly up-regulated by the treatment of MDV on14d.p.i(P<0.05),and the mRNA expression of COX2on22d.p.i was higher than on14d.p.i(P<0.05);On14and22d.p.i,the mRNA expression levels of PTGER4,HPGDS and GSTA3in the treatment group were significantly lower than that of the control group(P<0.05),and the mRNA expression level of PTGR1in the treatment group was significantly higher than that of the control group(P<0.05);The mRNA expression level of Ex-FABP in the treatment group was significantly lower than that of the control group on14d.p.i(P<0.05),but it was higher than that of the control group on22d.p.i.(P<0.05).In conclusion,the six immune-related genes in erythrocytes have significant immune responses after MDV infection,and erythrocytes can participate in the immunomodulatory effects in the process of MDV infection.
