运输应激对蛋鸡卵巢iNOS活性、NO含量及性激素受体基因表达的影响 被引量:3

摘要 为了研究运输应激对蛋鸡卵巢诱导型一氧化氮合酶(iNOS)活性、一氧化氮(NO)含量及性激素受体基因表达的影响,选取40只32周龄体重相近健康的海兰褐蛋鸡,随机分为对照组和试验组(每组20只),对试验组施以60km/h 6h的道路运输。运输结束后从对照组和试验组随机选取6只处死,采集卵巢组织,采用一氧化氮合酶(NOS)测定试剂盒、NO测定试剂盒和考马斯亮蓝法蛋白质定量测试盒及RTqPCR方法测定上述指标。结果表明,试验组蛋鸡卵巢iNOS活性极显著高于对照组(P<0.01),试验组NO含量极显著高于对照组(P<0.01);试验组蛋鸡卵巢雌二醇受体(E2R)mRNA相对表达量极显著低于对照组(P<0.001),试验组孕酮受体(PR)mRNA相对表达量极显著低于对照组(P<0.001),试验组睾酮受体(AR)mRNA相对表达量极显著高于对照组(P<0.001)。说明运输应激显著影响蛋鸡卵巢iNOS活性、NO含量及性激素受体基因的表达,可能引起卵巢组织发生炎症反应,引起卵巢组织损伤,从而影响生殖激素水平,可能导致蛋鸡生产性能下降。 The objective of this study was to investigate the effects of transport stress on iNOS activity,NO content and gonadal hormone receptor mRNA expressions in laying hens′ovaries.A total of4032weeks of similar weight Hyline Brown hens were randomly divided into control group and experimental group with20hens each respectively.The experimental group was imposed to6h road transport at the speed of60km/h.After transporting,6hens were randomly selected from the control group and the experimental group.The ovarian tissues were collected and the related indexes were measured by NOS,NO and Coomassie(bradford)protein assay kits and qRT-PCR.The results showed iNOS activity in the ovary of the experimental group was significantly increased than that of control group(P<0.01),NO content of the experimental group was significantly increased than that of control group(P<0.01);E2R mRNA relative expression of the experimental group was significantly decreased than that of control group(P<0.001),PR mRNA relative expression of the experimental group was significantly decreased than that of control group(P<0.001),AR mRNA relative expression of the experimental group was significantly increased than that of control group(P<0.001).In conclusion,transport stress has significantly effects on iNOS activity,NO content and gonadal hormone receptor mRNA expressions in laying hens′ovary which may cause ovarian tissue inflammation reaction and injury,thus affect reproductive hormone levels which result in a decline in reproduction performance of laying hens.
