摘要
目的研究黄秋葵醇提物对2型糖尿病小鼠胰岛素抵抗的影响。方法昆明小鼠尾静脉注射链脲佐菌素(STZ) 60 mg/kg 6周后,检测小鼠的空腹血糖(FBG),小鼠以高脂饲料进行喂养。将FBG≥11. 1 mol/L的小鼠随机分为模型组,二甲双胍组(0. 7 g/kg),黄秋葵醇提物0. 2、0. 4、0. 8 g/kg组,每组10只。另设10只昆明小鼠为正常组,给予普通饲料喂养。灌胃给药14 d,每天1次。末次给药前禁食不禁水12 h,检测小鼠FBG;末次给药1 h后摘眼球法取血,分离血清。放射免疫测定法测定血清中空腹胰岛素(FNS)含量,并计算其胰岛素敏感指数(ISI);酶联免疫法测定血清中肿瘤坏死因子-α(TNF-α)、白介素-6 (IL-6)、瘦素(Leptin)、脂联素(APN)、游离脂肪酸(FFA)的含量。蛋白免疫印记法检测肝脏组织中PPARα、PEPCK的蛋白表达水平。结果与模型组比较,黄秋葵醇提物治疗组的小鼠FBG均降低(P <0. 05),ISI、APN含量升高(P <0. 05);血清中TNF-α、IL-6、Leptin、FFA含量均降低(P <0. 05);肝脏组织中PPARα的表达增加,PEPCK的蛋白表达减少(P <0. 05)。结论黄秋葵醇提物对2型糖尿病小鼠具有降低血糖、改善胰岛素抵抗的作用,可能与通过调节PPARα、PEPCK蛋白的表达,影响血清中的炎症因子、FFA、Leptin、APN的表达有关。
Objective To investigate the effect of ethonal extracts of Abelmoschus esculentus Moench on insulin resistance of type2diabetes mellitus(T2DM).Methods The Kunming mice were injected with streptozotocin(STZ60mg/kg)through tail vein and given high-fat diet.After6weeks,the mice were considered as T2DM mice model with fasting blood glucose(FBG)>11.1mol/L.The T2DM mice were divided into following groups:model control group,metformin group(0.7mg/kg),ethonal extracts of Abelmoschus esculentus Moench groups(0.2,0.4and0.8mg/kg),10mice per group.Other10normal mice were considered as normal group.Mice were administered once a day for14days.Before last administration,the mice were fasting over night for12hours.One hour after administration,the serum and liver tissues were collected.The FBG,fasting insulin(FNS)were detected and insulin sensitivity index(ISI)was calculated.The levels of tumor necrosis factor-alpha(TNF-α),interleukin-6(IL-6),leptin,adi-ponectin(APN),free fatty acids(FFA)were determined.The protein expressions of PPARαand PEPCK in liver tissues were detected by western blot.Results Compared with model control group,the FBG was significantly decreased by ethonal extracts of Abelmoschus esculentus Moench(P<0.05),since,the ISI and APN were increased(P<0.05).The serum levels of TNF-α,IL-6,Leptin and FFA were reduced by ethonal extracts of Abelmoschus esculentus Moench(P<0.05).The protein expression of PPARαwas up-regulated,while the PEPCK was downregulated(P<0.05).Conclusion The ethonal extracts of Abelmoschus esculentus Moench can decrease the FBG and improve the insulin resistance.The underlying mechanism may be related with the regulation of PPARα,PEPCK protein expressions and the expressions of inflammatory factors,FFA,Leptin and APN.
