体外抗氧化试验筛选抗氧化肽的方法比较 被引量:1

摘要 为获得筛选抗氧化肽的快速、准确和重复性高的测定方法,利用具有抗氧化活性的谷胱甘肽对文献报道中常用的7种生物化学体外抗氧化实验进行筛选,对筛选后的方法在采用不同试验人员进行验证,而后对验证重复性比较好的方法再用合成的具有抗氧化活性的肽568 (Pro-Phe-Thr-Arg-Asn-Tyr-Tyr-Val-Arg-Ala-Val-Leu-HisLeu)、肽580(Trp-Asn-Trp-Ala-Asp)、肽567(Val-Tyr-Gln-Phe-Leu)和肽467(Pro-His-Cys-Lys-Arg-Met)进行确认,数据处理采用origin8.0和SPSS15.0进行。结果表明:常用的清除1,1-二苯基-2-三硝基苯肼(1,1-diphenyl-2-picrylhydrazyl,DPPH·)和超氧化物歧化酶(superoxide dismutase,SOD)这两种方法稳定性和重现性最佳,清除DPPH·和SOD可以作为抗氧化肽初步筛选的方法。 In order to obtain a rapid,accurate and reproducible assay for screening antioxidant peptides,screening of seven biochemical in vitro antioxidant experiments commonly used in the literature reports using glutathione with antioxidant activity,the method after screening was verified by different experimenters.Then,the method for verifying the reproducibility was confirmed by using the synthesized peptide 568(Pro-Phe-Thr-Arg-Asn-Tyr-Tyr-Val-Arg-Ala-Val-Leu-His-Leu),peptide 580(Trp-Asn-Trp-Ala-Asp),peptide 567(Val-Tyr-Gln-Phe-Leu)and peptide 467(Pro-His-Cys-Lys-Arg-Met)peptides having antioxidant activity,and the data processing was carried out using origin 8.0 and SPSS 15.0.The results were showed that the two methods of 1,1-diphenyl-2-picrylhydrazyl(DPPH·)and superoxide dismutase(SOD)are the best for stability and reproducibility.The removal of DPPH·and SOD can be used as the preliminary screening methods for antioxidant peptides.
