大蒜素对白念珠菌感染的免疫抑制小鼠外周血细胞因子的影响 被引量:1

摘要 目的 探讨大蒜素对白念珠菌感染的免疫抑制小鼠外周血细胞因子的影响。方法 BALB/c小鼠30只通过腹腔注射环磷酰胺100 mg/kg进行免疫抑制,后尾静脉接种2×107/ml白念珠菌菌液进行感染。将白念珠菌感染的免疫抑制小鼠随机分为免疫抑制组、大蒜素治疗组、生理盐水对照组。大蒜素治疗组灌服大蒜素、生理盐水对照组灌服等量生理盐水、免疫抑制组未做处理,共7 d。通过摘除眼球取血,收集正常小鼠、免疫抑制小鼠、大蒜素治疗组小鼠、生理盐水对照组小鼠外周血血清,ELISA测定IL-2、IFN-γ等Th1型和IL-4、IL-10等Th2型细胞因子的水平。结果 与正常小鼠比较,免疫抑制组、生理盐水对照组外周血IL-2、IFN-γ、IL-4、IL-10水平均降低,差异有统计学意义(P<0.05);大蒜素治疗组比免疫抑制组、生理盐水对照组明显增高(P<0.05),与正常小鼠相比差异无统计学意义(P>0.05);大蒜素治疗组外周血IL-4、IL-10比IL-2、IFN-γ增高更明显(P<0.05)。结论 大蒜素可提高白念珠菌感染的免疫抑制小鼠外周血细胞因子水平,可能通过此细胞因子网络而发挥一定的抗真菌感染作用。 Objective To investigate the effect of allicin on peripheral blood cytokines in immunosuppressed mice infected with candida albicans.Methods 60 BALB/c mice were immunosuppressed through intraperitoneally injected with cyclophosphamide 200 mg/kg,then inoculation of 2×107/ml candida albicans in the tail vein for infection.Immunosuppressive mice infected with candida albicans were randomly divided into immunosuppressive group,allicin treatment group,and normal saline control group.The allicin treatment group was given allicin,and the normal saline control group was given the same amount of normal saline for 7 days.Peripheral blood serum of normal mice,immunosuppressed mice,allicin treatment mice,and control mice were collected through removing the eyeball.The levels of Th1 type cytokines such as IL-2 and IFN-γ,and Th2 type cytokines such as IL-4 and IL-10 were measured by ELISA.Results Compared with normal mice,the levels of IL-2,IFN-γ,IL-4,and IL-10 in the peripheral blood of the immunosuppressive group and the normal saline control group decreased,with statistically significant differences(P<0.05);those of the allicin treatment group were significantly higher than those of the immunosuppressive group and the normal saline control group(P<0.05),but there were no statistically significant differences compared with normal mice(P>0.05).The levels of IL-4 and IL-10 in peripheral blood of allicin treatment group were significantly higher than the levels of IL-2 and IFN-γ(P<0.05).Conclusion Allicin can increase the level of peripheral blood cytokines in immunosuppressed mice infected with candida albicans,and may exert certain antifungal infection effects through this cytokine network.
