摘要 目的探讨肽基脯氨酰顺反异构酶1(Pin1)和细胞周期蛋白D1(Cyclin D1)在乳腺浸润性导管癌组织中的表达,分析Pin1和Cyclin D1表达与乳腺癌临床特征的相关性。方法选取2013年1月至2015年1月海南医学院第二附属医院保存的80例乳腺浸润性导管癌组织和40例癌旁正常乳腺组织标本,采用免疫组织化学法检测乳腺浸润性导管癌组织和正常乳腺组织中Pin1和Cyclin D1的表达,并分析Pin1和Cyclin D1表达与乳腺浸润性导管癌病理分级、TNM分期及淋巴结转移的相关性。结果乳腺浸润性导管癌组织中Pin1和Cyclin D1阳性表达率分别为56. 3%(45/80)、45. 0%(36/80),正常乳腺组织中Pin1和Cyclin D1阳性表达率分别为12. 5%(5/40)、20. 0%(8/40);乳腺浸润性导管癌组织中Pin1和Cyclin D1阳性表达率显著高于正常乳腺组织(χ~2=21. 001、7. 170,P <0. 05)。Pin1表达与乳腺浸润性导管癌病理学分级及淋巴结转移有关(χ~2=10. 381、4. 085,P <0. 05),与TNM分期无关(χ~2=2. 544,P> 0. 05)。Cyclin D1表达与乳腺浸润性导管癌病理学分级有关(χ~2=7. 024,P <0. 05),与TNM分期及淋巴结转移无关(χ~2=3. 236、0. 191,P> 0. 05)。乳腺浸润性导管癌组织中Pin1与Cyclin D1表达呈正相关(χ~2=4. 363,P <0. 05)。结论 Pin1和Cyclin D1在乳腺浸润性导管癌组织中表达上调,其可能与乳腺浸润性导管癌的发生、发展有关。 Objective To investigate the expression of peptidyl-prolyl cis-trans isomerase 1(Pin1)and Cyclin D1 in invasive ductal carcinoma of breast,and to analyze the correlation between the expression of Pin1 and Cyclin D1 and the clinical characteristics of breast cancer.Methods Eighty cases of invasive ductal breast cancer and forty cases of normal breast tissue specimens in the Second Affiliated Hospital of Hainan Medical University from January 2013 to January 2015 were selected.The expression of Pin1 and Cyclin D1 in invasive ductal carcinoma and normal breast tissues were detected by immunohistochemistry.The correlation between Pin1 and Cyclin D1 expression and pathological grading,TNM stage and lymph node metastasis of invasive ductal carcinoma was analyzed.Results The positive expression rates of Pin1 and Cyclin D1 in breast invasive ductal carcinoma were 56.3%(45/80)and 45.0%(36/80),respectively;the positive expression rates of Pin1 and Cyclin D1 in normal breast tissues were 12.5%(5/40)and 20.0%(8/40),respectively;the positive expression rates of Pin1 and Cyclin D1 in breast invasive ductal carcinoma tissues were significantly higher than those in normal breast tissues(χ^2=21.001,7.170;P<0.05).The expression of Pin1 was correlated with pathological grading and lymph node metastasis of breast invasive ductal carcinoma(χ^2=10.381,4.085;P<0.05),but it was not related to TNM stage(χ^2=2.544,P>0.05).The expression of Cyclin D1 was correlated with pathological grading of invasive ductal carcinoma(χ^2=7.024,P<0.05),but it was not related to TNM staging and lymph node metastasis(χ^2=3.236,0.191;P>0.05).The expression of Pin1 was positively correlated with CyclinD1 expression in invasive ductal carcinoma(χ^2=4.363,P<0.05).Conclusion The expressions of Pin1 and Cyclin D1 are up-regulated in invasive ductal carcinoma of breast,it may be related to the occurrence and development of invasive ductal carcinoma of breast.
