PD-1/PD-L1在复发/难治经典霍奇金淋巴瘤中的应用进展 被引量:3

Advances in anti-PD-1/PD-L1 antibodies for the treatment of relapsed/refractory classical Hodgkin's lymphoma

摘要 经典霍奇金淋巴瘤(classical Hodgkin's lymphoma,cHL)通常预后较好,但复发/难治经典霍奇金淋巴瘤(relapsed/refractory classical Hodgkin's lymphoma,R/R cHL)患者仍难以获得长期缓解和生存。cHL的R-S细胞(Reed-Sternberg cells)存在9p24.1染色体异常,导致程序性死亡配体-1(programmed death ligand-1,PD-L1)和程序性死亡配体-2(programmed death ligand-2,PD-L2)过表达,与T细胞表面程序性死亡受体-1(programmed death-1,PD-1)结合,诱导免疫耐受。PD-1或PD-L1抗体可阻断PD-1/PD-L1通路,逆转免疫耐受,发挥抗肿瘤作用。目前已有多种抗PD-1/PD-L1抗体用于治疗R/R cHL,在单药及联合治疗时显示出良好的耐受性和治疗效果。本文对PD-1/PD-L1在R/R cHL治疗中的应用进展进行综述。 Classical Hodgkin's lymphoma(cHL)is a sort of hematopoietic and lymphoid malignancy usually with favorable prognosis.However,long-term remission and survival of relapsed/refractory classical Hodgkin's lymphoma(R/R cHL)represent persistent challenges.Reed-Sternberg cells of cHL harbor alterations in chromosome9p24.1,leading to the overexpression of programmed death ligand-1(PD-L1)and programmed death ligand-2(PD-L2),ligands of the programmed death-1(PD-1)immune checkpoint receptor on T cells,and evading anti-tumor immune responses.The anti-PD-1and anti-PD-L1antibodies block the PD-1/PD-L1interaction and have shown robust antitumor activity.To date,several anti-PD-1/PD-L1antibodies have been developed for the treatment of R/R cHL with substantial response rates and acceptable safety profiles.This review summarizes the advances of anti-PD-1/PD-L1therapy in R/R cHL.
