摘要
【目的】研究大豆苷元抑制人成骨样MG-63细胞分泌IL-6的分子机制。【方法】选用兼有两种雌激素受体(estrogen receptor,ER)亚型表达的人成骨样MG-63细胞作为细胞模型,应用RT-PCR、ELISA、荧光素酶报告基因检测及小RNA干扰技术,研究大豆苷元对人成骨样MG-63细胞分泌骨吸收调节因子IL-6的作用及其分子机制。【结果】大豆苷元可降低MG-63细胞IL-6的分泌,抑制mRNA表达和基因启动子的转录活性,上述作用能够被ER阻断剂ICI 182,780所阻断。小RNA干扰技术进一步证实大豆苷元对MG-63细胞IL-6表达的抑制作用主要由ERα介导。【结论】大豆苷元可以抑制人成骨样MG-63细胞分泌IL-6,其调节作用与ER通路有关,主要由ERα介导。
【Objective】To study the molecular mechanism of daidzein on inhibiting IL-6 secretion in human osteoblast-like MG-63 cells.【Methods】Human osteoblast-like MG-63 cells containing two estrogen-receptor(ER)isoforms were chosen as a suitable cell model for this study.RT-PCR,enzyme-linked immunosorbent assay(ELISA),luciferase reporter assay and small interfering RNA technology were performed to study the effect of daidzein on IL-6 secretion in osteoblast-like MG-63 cells and underlie its molecular mechanism.【Results】Daidzein obviously inhibited the IL-6 secretion,mRNA expression and IL-6 promoter activity in human osteoblastic MG-63 cells,which could be abrogated by ER antagonist ICI 182,780.By siRNA technology,it was further demonstrated that the inhibition of daidzein on IL-6 expression was mediated by ERα.【Conclusion】The effect of daidzein on IL-6 secretion in human osteoblast-like MG-63 cells is associated with ER pathway and mainly mediated by ERα.
