摘要
为探明煤矿区土壤微生物资源和生态分布特点,2016年10月从河北省邯郸市峰峰矿区采集冬青(Ilex chinensis)、小檗(Berberis kawakamii)和构树(Broussonetia papyrifera)根围土壤样品,利用磷脂脂肪酸(PLFA)法结合Sherlock微生物鉴定系统分析土壤微生物群落结构。结果表明:峰峰矿区植物根围土壤微生物PLFA种类丰富,其中以15∶0iso、16∶1ω7c、16∶0 10-methyl、18∶1ω9c和18∶1ω7c为优势PLFA;细菌为优势菌群。3种植物PLFA总量和微生物群落结构差异显著,表现为构树>小檗>冬青,其中14∶0anteiso和17∶1isoω9c为构树特有;18∶3ω6c为小檗特有;在冬青土壤未检测到15∶1isoω6c和18∶1ω5c等特征PLFA。RDA分析结果表明:微生物群落与土壤Zn、Mn、Pb、Cu、Cd元素及pH、全氮、磷酸酶和有机碳显著正相关,与速效磷显著负相关;土壤Cd、全氮、有机碳、速效磷和磷酸酶是土壤微生物PLFA的主要影响因子。冬青、小檗和构树的重金属富集能力较强,可作为矿区修复的理想植被。
To elucidate soil microbial resources and distribution in coal mine area,the rhizosphere soils of Ilex chinensis,Berberis kawakamii and Broussonetia papyrifera from Fengfeng mining area in Hebei Province,China were collected in October 2016.Soil microbial community structure in the rhizosphere of the three plant studied was analyzed by phospholipid fatty acid(PLFA)combined with Sherlock microbial identification system.The results showed that soil microbial PLFA species in Fengfeng Mining Area was rich,with.15∶0 iso,16∶1ω7c,16∶0 10-methyl,18∶1ω9c and 18∶1ω7c as the dominant PLFAs,and Bacteria had a maximum value.Total PLFA and soil microbial community structure in rhizosphere soil of different plants were significantly different,characterized by B.papyrifera>B.kawakamii>I.chinensis.14∶0 anteiso and 17∶1 isoω9c were unique in rhizosphere of B.papyrifera,18∶3ω6c only existed in the rhizosphere of B.kawakamii,and 15∶1 isoω6c and 18∶1ω5c were undetected in rhizosphere of I.chinensis.RDA indicated that soil microbial communities were significantly positively correlated with soil Zn,Mn,Pb,Cu,Cd,pH,total nitrogen,phosphatase and organic carbon,and was extremely negatively correlated to available phosphorous.The Cd,total nitrogen,total organic carbon,available phosphorus and phosphatase were the main influencing factors of soil microbial PLFA.Based on the ability to strongly accumulate heavy metals,three plant studied can be used to repair the mining area as ideal vegetation.
