病死猪堆肥降氨除臭微生物的筛选与鉴定 被引量:6

摘要 为了获取高效的病死猪堆肥降氨除臭微生物菌株,利用氨气选择性培养基定性初筛和硼酸吸收法复筛相结合的方法,分离筛选高效降氨微生物,并对其菌种分类进行了鉴定。从猪粪、土壤、病死猪堆肥等样品中共分离筛选获得7株对NH3具有降解效果的菌株,分别编号为YX-1、YX-2、YX-3、YX-4、YX-6、YX-7、YX-8。在30 d模拟堆肥周期中,菌株YX-3对氨气的去除效率最高,达56. 9%。根据形态学特征、生理生化特征及16S r DNA序列分析,该菌鉴定为暹罗芽孢杆菌(Bacillus siamensi)。通过单因素试验,确定菌株YX-3降氨除臭的最佳温度为30~40℃,最佳接种量为8%。菌株YX-3对H2S的去除能力显著低于对氨气的去除能力,说明该菌去除臭气成分的功能专一。筛选获得的菌株YX-3丰富了堆肥除臭微生物资源库,在农业环保领域具有潜在的应用价值。 In order to obtain deodorant microorganisms for dead-pig composting,effective strains were screened by combining NH 3 selective media and the boric acid absorption method and the phylogenetic position of YX-3 was determined.Seven strains with ammonia emission ability were sceened from pig compost,soil and dead-pig compost samples,labeles as YX-1、YX-2、YX-3、YX-4、YX-6、YX-7、YX-8.In a composting experiment of 30 d,the NH 3 emission rate of strain YX-3 was 56.9%,which was higher than other six strains.Based on the results of morphology,physio-biochemical tests and 16S rDNA sequence analysis,strain YX-3 was identified as Bacillus siamensi.The single-factor test was used to determine the optimum deodorant conditions of YX-3.The optimum culture temperature was 30~40℃and the best inoculation amount was 8%.Compared with the NH 3 emission rate,YX-3 showed lower H 2S emission ability,which indicated that efficient H 2S emission strains should be screened for high deodorizing effect.The result shows that Bacillus siamensi YX-3 has great potential application prospects in the field of agriculture and environment protection.
