增强AhR表达对高糖状态牙龈卟啉单胞菌脂多糖导致的巨噬细胞炎症反应的影响 被引量:2

摘要 目的:探讨增强芳香烃受体(AhR)对高糖状态牙龈卟啉单胞菌(Pg)脂多糖(LPS)导致的巨噬细胞炎症反应的影响。方法:将小鼠巨噬细胞RAW264.7接种在高糖培养基中,并转染特异性增强AhR表达的重组质粒,稳定转染后将细胞在高糖状态加入Pg LPS刺激。用免疫印迹实验检测细胞的AhR及炎症反应相关蛋白Notch1、Jagged1、iNOS的表达,并用ELISA检测细胞培养上清液中的促炎因子TNFα、IL1β浓度。结果:在受高糖加Pg LPS刺激后,与未增强AhR表达的对照组细胞相比,增强AhR表达的细胞Notch1、Jagged1、iNOS、TNFα、IL1β蛋白表达量减少(P<0.05)。结论:增强AhR表达,可抑制高糖状态Pg LPS导致的巨噬细胞炎症反应,该抑制作用可能与AhR调节Notch通路有关。 Objective:To investigate the effects of enhanced expression of aryl hydrocarbon receptor(AhR)on the inflammatory response of macrophages induced by Porphyromonas gingivalis lipopolysaccharide(Pg LPS)under high glucose conditions.Methods:RAW264.7murine macrophages were inoculated in the medium with high glucose and were transfected with recombinant plasmids to enhance AhR expression.Then,Pg LPS was added to the medium with high glucose to induce inflammatory response.Western blotting was chosen to examine the expression of AhR,Notch1,Jagged1and iNOS in macrophages,and ELISA was used to detect the concentration of TNFαand IL1βin the culture supernatant.Results:Compared with the cells without enhanced AhR expression,the cells overexpressing AhR exhibited less expression of Notch1,Jagged1,iNOS,TNFα,and IL1βunder the condition of Pg LPS stimulation superimposed on high glucose(P<0.05).Conclusion:Increased AhR expression can inhibit the inflammatory response of macrophages induced by Pg LPS under high glucose conditions,and the suppresive effect may be associated with the inhibition of Notch signaling by AhR.
