摘要
目的探讨五味子乙素对过氧化氢(H_2O_2)所诱发的晶状体上皮细胞氧化损伤的影响。方法摘取SD大鼠晶状体后随机分为空白对照组、五味子乙素组(5 mmol/L五味子乙素)、氧化损伤组(1 mmol/L H_2O_2)、氧化损伤+五味子乙素组(1 mmol/L H_2O_2+5 mmol/L五味子乙素)离体培养24 h,显微镜下观察晶状体混浊情况,透射电子显微镜观察晶状体上皮细胞超微结构改变,Western-blot法检测晶状体上皮细胞中血红素加氧酶-1(heme oxygenase 1,HO-1)、核转录因子红系2相关因子2(nuclear factor-erythroid 2-related factor 2, Nrf-2)的蛋白表达。结果晶状体透明度观察结果显示,氧化损伤组晶状体透明度明显下降,而氧化损伤+五味子乙素组晶状体混浊情况明显轻于氧化损伤组;透射电子显微镜结果显示,空白对照组及五味子乙素组晶状体上皮细胞胞膜完整连续,核仁形态规则;氧化损伤组晶状体上皮细胞呈弥漫性改变,胞膜残缺不全,部分核碎裂;氧化损伤+五味子乙素组多数晶状体上皮细胞形态改变轻微,可看到连续的核膜结构。氧化损伤组大鼠晶状体上皮细胞中HO-1及Nrf-2含量明显低于空白对照组及五味子乙素组(P<0.05)。氧化损伤+五味子乙素组大鼠晶状体上皮细胞中HO-1及Nrf-2含量高于氧化损伤组(P<0.05)。结论五味子乙素对晶状体上皮细胞氧化损伤具有抑制作用,其作用机制可能与上调HO-1及Nrf-2表达有关。
OBJECTIVE To discuss the effects of Schisandrin B on oxidative damage in lens epithelial cells induced by H2O2.METHODS The lens of SD rats were gathered and randomly divided into blank control group,schisandrin B group(5mmol/L schisandrin B),oxidative damage group(1mmol/L H2O2)and oxidative damage plus schisandrin B group(1mmol/L H2O2+5mmol/L schisandrin B).They were cultured in vitro for24h and lens opacity were detected by microscope.Transmission electron microscope was used to detect lens epithelial cell ultrastructure change,Western blot method was used to observe protein expression of heme oxygenase(HO-1)and nuclear factor-erythroid2-related factor2(Nrf-2).RESULTS Lens transparency observation results showed that lens transparency in oxidative damage group decreased obviously,and lens opacity in oxidative damage plus Schisandrin B group were obviously lighter than that in oxidative damage group;Electron microscopy results showed full continuous lens epithelial cell membrane and regular shape nucleoli in the blank control group and Schisandrin B group;Lens epithelial cells in oxidative damage were found in diffuse change,cell membrane was incomplete,some nuclear was in fragmentation;most of the lens epithelial cells in oxidative damage plus Schisandrin B group changed slightly,continuous nuclear membrane structure was observed.The HO-1and Nrf-2level in the oxidative damage group were significantly lower than that in control group and Schisandrin B group(P<0.05).The HO-1and Nrf-2level in oxidative damage plus Schisandrin B group were higher than that in oxidative damage group(P<0.05).CONCLUSIONS Schisandrin B inhibits oxidative damage of lens epithelial cells,which might relate to increased expression of HO-1and Nrf-2.
