摘要
目的观察益精杞菊地黄颗粒对慢性高眼压大鼠视网膜神经节细胞(RGCs)凋亡的影响。方法选用健康雄性的SD大鼠60只,随机分为5组,空白组、模型组、高剂量中药组、中剂量中药组、低剂量中药组,每组各12只。除空白组,其余4组均采用烧灼巩膜上静脉法,建立慢性高眼压模型。干预1个月后,分别采用眼压测量观察眼压变化,HE染色观察视网膜病理学改变,Tunel法检测RGCs凋亡情况。结果 (1)眼压测量:造模后大鼠眼压明显高于造模前和空白组眼压水平,差异有统计学意义(P<0.01),模型组与各中药组比较无统计学意义(P>0.05)。用药后1个月,模型组及各中药组与造模前、空白组比较有统计学意义(P<0.01),而与造模后用药前比较,无统计学意义(P>0.05),但各中药组眼压有所下降。各用药组间比较均无统计学意义(P>0.05),与模型组比较均无统计学意义(P>0.05)。(2)HE染色显示,与空白组不同,模型组损伤明显;中剂量中药组较模型组损伤改善。模型组RGCs层细胞数量较正常组显著减少(P<0.01);高、中剂量中药组细胞数量高于模型组(P<0.01);低剂量中药组与模型组比较无统计学意义(P﹥0.05)。(3)Tunel检测显示,各治疗组RGCs层细胞凋亡较模型组减少,中剂量中药组细胞凋亡最少。空白组与模型组比较,差异有统计学意义(P<0.01),中剂量中药组凋亡率较模型组有统计学意义(P<0.01),高、低剂量中药组凋亡率与模型组比较,无统计学意义(P>0.05)。结论益精杞菊地黄颗粒剂对慢性高眼压大鼠RGCs的凋亡有明显的保护作用。
OBJECTIVE To observe the effect of Yijing Qiju Dihuang granule on retinal ganglion cells(RGC)apoptosis of chronic high intraocular pressure rats.METHODS Sixty male Sprague-Dawley rats were divided into5groups randomly:blank group,model group,high dose Chinese medicine group,medium dose group and low dose group,with12rats in each group.In addition to the blank group,rats in the other4groups were induced into chronic ocular hypertension model.All groups were intervened for one month.We used tonometry to observe the changes of intraocular pressure of every group.The pathological changes of retinal tissues of rats in every group were detected by hematoxylin and eosin(HE)staining.The RGCs apoptosis rates of every group were observed by Tunel detection.RESULTS1)Tonometry:immediate after modeling,intraocular pressure of rats in groups other than blank group increased dramatically and they were also higher than the result of blank group(P<0.01).Comparison between model group and Chinese medicine groups showed no statistical differences(P>0.05).One month after intervention,results of Chinese medicine groups and model group varied significantly and they were also different from blank group(P<0.01).However,they showed no difference with the result between modeling and intervention(P>0.05).The intraocular pressure of medication groups decreased to some extent.There was no significant difference between each medication group and model group(P>0.05).(2)The HE staining showed obvious damage in model group while not in blank group.Results of middle dose group were better than model group.The RGCs apoptosis of model group was lower than blank group(P<0.01).Cells count in high dose group and medium dose group were higher than model group(P<0.01).But it did not happen in low dose group(P>0.05).(3)Tunel test showed that the RGC apoptosis of intervention groups were lower than model group and result of medium dose group was the most superior.The RGCs apoptosis rate of model group was significantly higher than the blank group(P<0.01).While result of medium dose Chinese medicine group was better than model group(P<0.01).And results of high and low dose group were not statistically different to model group(P>0.05).CONCLUSIONS Yijing Qiju Dihuang granule had obvious protective effect on retinal ganglion cells apoptosis in chronic high intraocular pressure rats.
