摘要 目的探究血液标本在不同保存时间与保存温度条件下,采用Sysmex XN 1000血液分析仪测定血液标本血细胞各项参数结果的差别。方法采用Sysmex XN 1000血液分析仪测定120份血液标本,将其置于不同温度下保存并测定不同保存时间下血细胞各项参数。120份标本分为3组,每组各40份,第1组:置于-20℃冰柜冷冻,第2组:置于4℃冰箱保存,第3组:置于20℃室温保存,分别检测即刻、1、3、7d的血细胞参数。结果 -20℃冷冻条件下,保存1、3、7d时红细胞、白细胞数量逐渐降低,血红蛋白水平无明显变化,血小板在保存1~3d无明显变化,在3~7d逐渐升高,中性粒细胞保存1d后数量逐渐降低,淋巴细胞保存1d后数量逐渐升高,单核细胞保存1d后数量逐渐降低,嗜酸性粒细胞、嗜碱性粒细胞数量无明显变化。4℃低温保存条件下,保存1、3、7d时红细胞、血小板数量无明显变化,保存1d内白细胞数量无明显变化,保存3~7d白细胞数量稍微降低,保存1~3d血红蛋白水平无明显变化,保存3~7d血红蛋白水平逐渐升高。20℃室温保存条件下,保存1~3d红细胞数量无明显变化,保存3~7d红细胞数量开始有所升高,保存1、3、7d时白细胞数量持续升高,保存1、3、7d时血小板数量无明显变化,保存1~3d血红蛋白水平无明显变化,保存3~7d血红蛋白水平开始逐渐升高。结论血常规标本最适合在4℃低温条件下保存,标本保存1d时红细胞、白细胞、血小板和血红蛋白水平无明显变化,但白细胞分类结果有明显变化;保存3~7d白细胞数量稍微降低,血红蛋白水平逐渐升高。因此,临床上采集标本后应立即送检。 Objective To explore the effects of different storage temperature and time on the results of blood analysis measured by Sysmex XN1000blood and yzer.Methods A total of120blood samples were determined by XN1000hematology analyzer.The samples were stored at different temperature and the parameters of blood cells were determined at different storage time.A total of120blood specimens were divided into three groups and stored in different temperatures and times.The blood cells were examined by XN1000automated hematology analyzer.First group,40blood samples were stored in-20℃and analyzed at immediately1,3and7day(s);second group,40blood samples stored at4℃and analyzed at immediately1,3and7day(s);third group,40blood samples stored at20℃and analyzed at immediately1,3and7day(s).Results The number of red blood cells and white blood cells decreased gradually after storage for1,3and7day(s)at-20℃,but there was no significant change in hemoglobin.The platelets did not change significantly at1-3day(s),but increased gradually at3-7day(s),and the number of neutrophils decreased gradually after1day storage.The number of lymphocyte increased gradually after1day storage,the number of monocyte decreased gradually after1day storage,and the number of eosinophils and basophils did not change significantly.The number of red blood cells and platelets did not change significantly after1,3and7day(s)of cryopreservation at4℃,and the number of white blood cells did not change significantly within1day,while the number of white blood cells decreased slightly between3and7day(s).The amount of hemoglobin did not change significantly after storage for1-3day(s),but increased gradually after storage for3-7day(s).At room temperature,the number of red blood cells remained almost unchanged for1-3day(s),and began to increase after3-7day(s).After1,3and7day(s)of storage,the number of white blood cells continued to increase;after1,3and7day(s)of storage,the number of platelets remained almost unchanged;after1-3day(s)of storage,the number of hemoglobin remained almost unchanged;and after3-7day(s),the number of hemoglobin began to increase gradually.Conclusion Blood routine samples are most suitable for cryopreservation at4℃.The results of red blood cells,white blood cells,platelets and hemoglobin stored for1day basically have no obvious changes,but the classification results of white blood cells have obvious changes.3-7day(s),the number of white blood cells decreased slightly,and the number of hemoglobin increased gradually.Therefore,clinical samples should be sent immediately after collection.
