宁波舟山港域内5种病毒监测与遗传进化特征分析

摘要 为探索宁波舟山港域内发热伴血小板减少综合征(SFTS)的发病率与病死率明显高于省内其他地区的原因,为今后SFTS的检疫、防控与诊治提供科学依据。对发热伴血小板减少综合征病毒(SFTSV)及4种类SFTSV病毒采用荧光RT-PCR及间接免疫荧光法用于疑似病例血清样本中病毒的检测,并对确认阳性的样本进行病毒分离、序列测定与基因进化分析。在蜱、螨、鼠传播媒介中SFTSV及4种类SFTSV病毒的检测结果均为阴性,在180例疑似病例血样中仅检出SFTSV,阳性率为51.11%(92/180)。样本病毒分离阳性率为72.31%(47/65)。所分离SFTSV病毒的序列分析结果显示出A与B两个基因型及5个进化分支,其中A型占25.53%、B型占74.47%。当前流行于宁波舟山港域内的SFTSV病毒仅为SFTSV且以B基因型为主。B型SFTSV病毒在国内罕见,在遗传进化上具有相对独立的进化特征。 To find out the reason of the status that the incidence and mortality of Severe Fever with Thrombocytopenia Syndrome(SFTS)in Ningbo-Zhoushan port area were significantly higher than those in other area of Zhejiang province,and provide scientific evidence for future inspection and quarantine,diagnosis and treatment of SFTS.Viral detection in samples were carried out by both established real-time fluorescent quantitative RT-PCR of SFTSV and SFTSV like viruses,and indirect immunofluorescence assays.Virus isolation,sequencing and gene evolution analysis of confirmed positive samples were done as well.The tested results of SFTSV and SFTSV like viruses showed negative in all detected medium.Only SFTSV viruses were found in the blood of 180 clinical suspected cases,and the positive rate of SFTSV was 51.11%(92/180).The positive rate of virus isolation was 72.31%(47/65).Sequence analysis showed that there were 5 evolutionary branches of A and B genotypes.A type accounted for 25.53%and B type accounted for 74.47%.The pathogen of SFTS prevailing in Ningbo-Zhoushan Port area is SFTSV currently which main genotype is B.The B genotype virus has relatively independent evolutionary features in genetic evolution.It is a rare genotype in China.
