拉莫三嗪胶束经鼻给药入脑后对IL-6水平的影响

摘要 目的通过检测拉莫三嗪mPEG-PLA聚合物胶束经鼻入脑后对大鼠IL-6细胞因子的影响,研究胶束制剂入脑后是否会引起免疫应答或炎症反应。方法各组大鼠分别鼻腔滴注给予拉莫三嗪胶束或生理盐水,采用ELISA法测定给药30 min和90 min后,大鼠脑组织、颈深淋巴结、颈静脉血和脑脊液IL-6的质量浓度。结果拉莫三嗪胶束鼻腔给药30 min后,脑组织及颈深淋巴结IL-6的水平较空白组明显提高,90 min后又恢复至正常水平;血液及脑脊液的IL-6水平在90 min内与生理盐水组比较差异无统计学意义。结论拉莫三嗪胶束入脑后能引起脑组织IL-6水平的上升,这个过程可能激活了免疫应答反应,但未引起机体炎症反应。 Objective To investigate the effect of lamotrigine mPEG-PLA polymer micelle on cytokine IL-6levels by nasal administration and study whether the micell would activate an immune response or cause an inflammation response.Methods The corresponding groups of rats were dosed with lamotrigine mPEG-PLA polymer micelle or saline solution by nasal administration,followed by determination of cytokine IL-6levels of brain tissue,deep cervical lymph nodes,cerebrospinal fluid,cervical venous blood by using ELISA method after 30 min and90min.Results The levels of IL-6in brain tissue and deep cervical lymph nodes had an obvious improvement comparing with control group within30min,and then returned to normal level after 90 min.In contrast,the levels of IL-6 in blood and cerebrospinal fluid had no significant difference within90min compared with that in saline group.Conclusion Nasal administration would induce a rapid increase of IL-6 levels in the brain tissue and deep cervical lymph nodes,which may activate the immune responses but not cause inflammation.
