摘要
目的通过本市848例临床疑似及确诊患者标本的DNA微阵列法检测,了解我市结核分枝杆菌的耐药情况,为临床诊断和治疗提供依据。方法采用结核分枝杆菌耐药基因检测试剂盒检测利福平和异烟肼耐药相关基因,采用加入0.5%N-乙酰-L-半胱氨酸优选液化和延长液化时间的方法,对实验进行优化。结果848份标本中共检出结核分枝杆菌571例(67.33%),结核分枝杆菌的总耐药率为20.02%,MDR-TB占结核分枝杆菌的10.68%。结论该市结核分枝杆菌耐药情况不容乐观。DNA微阵列法检测结核分枝杆菌的耐药基因能快速高效的为临床诊断和治疗提供依据,对实验的优化能提高耐药基因检出率。
Objective According to testing 848 cases of clinically suspected and confirmed patients in our city with the method of DNA microarray,to comprehend the resistance to mycobacterium tuberculosis in our city and provide a basis for clinical diagnosis and treatment.Methods The drug resistance gene detection kit of tuberculosis is used to test the genes related to the resistance of rifampicin and isoniazid and 0.5%n-acetyl-l-cysteine is used to get preferred liquefaction and extended liquefaction time,which make this experiment more optimized.Results There are 571 cases of mycobacterium tuberculosis(67.33%)in 848 cases,the total drug resistance rate of mycobacterium tuberculosis is 20.02%,MDR-TB accounts for 10.68%of mycobacterium tuberculosis.Conclusion The drug resistance of mycobacterium tuberculosis in our city is not optimistic.DNA microarray can detect drug resistance genes of mycobacterium tuberculosis rapidly and efficiently and provide evidence for clinical diagnosis and treatment,the optimization of the experiment can improve the detection rate of drug-resistant genes.
