摘要
采用FISH(荧光原位杂交)技术,分析普通小麦贵紫麦1号染色体的FISH核型特点,为贵紫麦1号在小麦新品种选育上的应用提供参考。结果表明,贵紫麦1号包括21对染色体,pAs 1红色探针信号主要分布在A组和D组染色体上,pSc 119.2-1绿色探针信号则主要分布在B组染色体上;根据贵紫麦1号染色体上这2种探针的分布特征,可以准确辨识每条染色体。贵紫麦1号与硬粒小麦(AABB)在2A、5A、2B、3B及5B染色体上表现出pSc 119.2-1信号分布差异,与节节麦(DD)在染色体1D、4D及5D上也存在信号分布差异,与中国春(AABBDD)在2A、5A、6B、1D及7D染色体上的信号也各有不同,说明DNA重复序列在不同小麦材料之间具有多态性。
FISH(Fluorescence in situ hybridization)technique was applied to analyze FISH patterns of chromosomes in Triticum aestivum L.(Guizimai No.1),which would provide reference for the application of this variety in the breeding of new wheat varieties in the present study.The results showed that Guizimai No.1 included 21 pairs of chromosomes,the pAs 1 red probe signals were mainly distributed on chromosomes of A and D genome,while the pSc 119.2-1 green probe signals were mainly distributed on the chromosomes of B genome.According to the distribution characteristics of these two probes on Guizimai No.1 chromosomes,each chromosome could be identified accurately.Generally,The differences in signal distribution of pSc 119.2-1 was showed between chromosomes 2 A,5 A,2 B,3 B and 5 B of Guizimai No.1 and durum wheat(AABB),and there was also a signal distribution differences on chromosomes 1 D,4 D and 5 D between Ae.Tauschii(DD),and the signal on chromosomes 2 A,5 A,6 B,1 D and 7 D were different from those of Chinese Spring(AABBDD).The results indicated that DNA repeat sequences had polymorphisms between different wheat materials.
