醒脾解郁法对抑郁大鼠肝脏能量代谢的调控机制研究 被引量:17

摘要 目的观察抑郁大鼠模型肝脏线粒体能量代谢特征及其与抑郁症行为学指征变化的动态相关性,并揭示以益气健脾为主的醒脾解郁方调节情志的效应机制。方法采用慢性不可预见性温和应激方法应激6周,应激同时灌胃给药,糖水偏爱实验、强迫游泳行为学评价作为造模成功标志,肝脏取材,并使用化学法检测线粒体损伤指标(呼吸链复合体Ⅰ、Ⅱ、Ⅲ、Ⅳ含量)和能量指标(ATP、Na-K-ATP酶、Ca-Mg-ATP酶含量)。结果检测模型大鼠肝组织线粒体指标、能量指标发现:应激2周后,与正常组相比,模型组肝组织内呼吸链复合体Ⅲ含量显著降低(P <0. 05),与模型组比较,各治疗组未见明显改善(P> 0. 05);应激6周后,与正常组相比,模型组肝组织内呼吸链复合体Ⅰ、Ⅲ、Ⅳ和ATP、Ca-Mg-ATP酶含量减少(P <0. 05),与模型组比较,中药高剂量组可显著增加肝组织内呼吸链复合体Ⅰ、Ⅲ、Ⅳ和ATP、Ca-Mg-ATP酶的含量(P <0. 05),西药舍曲林组可显著增加肝组织内呼吸链复合体Ⅳ含量(P <0. 05)。结论醒脾解郁方可以明显改善抑郁大鼠肝脏线粒体能量代谢功能,发挥抗抑郁效应。 Objective To observe the energy metabolic characteristics of mitochondria in the liver of depressed rat model and its dynamic correlation with the changes of behavioral indications of depression,and to reveal the mechanism of regulating emotion by Xingpi Jieyu decoction which mainly benefits qi and invigorates spleen.Methods The method of chronic unpredictable mild stress was applied for6weeks with sugar water administration.The syrup preference experiment and forced swimming behavioral evaluation were used as the signs for successful modeling.The liver was taken and the mitochondrial damage index(respiratory chain complexⅠ,Ⅱ,Ⅲ,Ⅳcontent)and energy index(ATP,Na-K-ATPase)were detected by chemical method,Ca-Mg-ATPase content).Results The mitochondrial index and energy index of the liver tissues of the model rats were detected.After2weeks of stressing,the content of respiratory chain complexⅢin the liver tissues of the model group was significantly reduced compared with the normal group(P<0.05),and there was no obvious improvement among other treatment groups compared with the normal group.After6weeks of stressing,compared with normal group,the contents of respiratory chain complexⅠ,Ⅲ,Ⅳand ATPase,Ca-Mg-ATP in the liver tissue of model group were decreased(P<0.05).Compared with model group,the contents of respiratory chain complexⅠ,Ⅲ,Ⅳand ATP and Ca-Mg-ATPase in the liver tissue were increased in the high Chinese medicine dose group(P<0.05),and the content of respiratory chain complexⅣin the liver tissue was significantly increased in the sertraline group(P<0.05).Conclusion Xingpi Jieyu decoction can significantly improve the energy metabolism function of liver mitochondria in depressed rats,which has an anti-depression effect.
