基于关联规则分析熟地黄配伍规律对防治骨质疏松症的启示

Enlightenmenton compatibility rules of Radix Rehmanniae Praeparata in the prevention and treatment of osteoporosis based on association rules

目的探讨中国古代方剂中熟地黄的配伍规律并为临床防治骨质疏松症提供新的思路与方法。方法利用Excel的筛选功能搜集中国科技资源共享网国家人口与健康科学数据平台中国方剂数据库中所有含熟地黄的方剂,并对药物名称规范化处理,分别在SPSS、Weka、Pajek统计分析软件中进行频次统计、关联规则挖掘、关联规则可视化处理。结果共搜集到487首熟地黄方,累计使用频率在50%以上的中药有熟地黄、当归、白芍、川芎、茯苓、白术、甘草、人参等24种,以益气补血为主;熟地黄的使用剂量多集中在2两以内,而以1两以内为多,其中1两以内又以1～2钱,3～4钱,5～6钱为主;关联规则中二项药物关联置信度较高的是山茱萸—熟地黄、泽泻—熟地黄、延胡索—当归等;三项药物关联置信度较高的是当归—茯苓—川芎、山药—山茱萸—熟地黄、山茱萸—山药—熟地黄等;四项药物关联置信度较高的是川芎—白芍—熟地黄—当归、当归—白芍—熟地黄—川芎、白术—茯苓—熟地黄—人参等。关联规则可视化处理显示,熟地黄常以四物汤、八珍汤、六味地黄汤的形式被遣用。结论运用熟地黄益气补血与填精益髓并重防治骨质疏松症可能是重要的或有效的治则治法。

Objective To investigate the compatibility rules of the Radix Rehmanniae Praeparata in ancient formulas and propose a new therapeutic principle in treating osteoporosis.Methods The Radix Rehmanniae Praeparata related formulas were collected from Chinese formulas database,Chinese medicine scientific data center of information institute,China Academy of Chinese Medical Science using Excel,then their names were standardized so as to be calculated and analyzed by SPSS,Weka and Pajek.In this paper,SPSS was exploited for descriptive analysis.Association rules were calculated by Weka and visualized by Pajek.Results A total of487formulas were found,and24kinds of TCM herbs with over50%of the utilization rate.They were Radix Rehmanniae Praeparata,Radix Angelicae Sinensis,Radix Paeoniae Alba,Rhizoma Ligustici Chuanxiong,Poria,Rhizoma Atractylodis Macrocephalae,Radix Glycyrrhizae,Radix Ginseng,etc.,which were mainly for supplementing qi and replenishing blood;Dosage of the Radix Rehmanniae Praeparata was mostly less than2liang,however,mostly within1liang,among which1~2qian,3~4qian,and5~6qian accounted the most.The top3confident-outcomes of association rules association rules were as follow:In2-herbs-associated group,they were Fructus Corni-Radix Rehmanniae Praeparata,Rhizoma Alismatis-Radix Rehmanniae Praeparata,Rhizoma Corydalis-Radix Angelicae Sinensis;In3-herbs-associated group,they were Radix Angelicae Sinensis-Poria-Rhizoma Ligustici Chuanxiong,Rhizoma Dioscoreae-Fructus Corni-Radix Rehmanniae Praeparata,Fructus Corni-Rhizoma Di-oscoreae-Radix Rehmanniae Praeparata;In4-herbs-connected group,they were Rhizoma Ligustici Chuanxiong-Radix Paeoniae Alba-Radix Rehmanniae Praeparata-Radix Angelicae Sinensis,Radix Angelicae Sinensis-Radix Paeoniae Alba-Radix Rehmanniae Praeparata-Rhizoma Ligustici Chuanxiong,Rhizoma Atractylodis Macrocephalae-Poria-Radix Rehmanniae Praeparata-Radix Ginseng.Association rules visualization showed that Radix Rehmanniae Praeparata was mostly used in Siwu decoction,Bazhen decoction and Liuwei Dihuang decoction.ConclusionIt might be an important or efficient principle to lay equal stress on enriching the kidney essence and tonifying qi and replenishing blood in treating osteoporosis by Radix Rehmanniae Praeparata.
