摘要
目的:探讨小鼠脾生后的形态学变化。方法:采用H-E染色观察小鼠生后脾的形态学变化,Giemsa染色观察造血细胞的变化,PAS染色观察巨核细胞的变化,CD36免疫组织化学检测巨核细胞在脾的表达和变化。结果:小鼠脾P1d红髓较多,P5d出现白髓,边缘区及边缘窦也逐渐形成,于P31d接近正常脾结构。巨核细胞多聚集成群,近被膜、血管及脾索周围多见。免疫组织化学显色可区分出颗粒型巨核细胞、产血小板型巨核细胞和巨核细胞裸核3种类型。颗粒型与裸核型大致呈增长趋势,产血小板型的变化趋势同前两者相反,三者随脾成熟后数量趋于平稳。结论:小鼠脾于P31d接近成年,且终身具有造血能力。CD36免疫组织化学显色可将巨核细胞分为颗粒型、产血小板型和裸核型。
To explore the morphological changes of mice spleen during postnatal days.Methods:The morphological changes of spleen was evaluated by H-E staining,the changes of hematopoietic cells was studied by Giemsa staining and the changes of megakaryocyte by PAS staining.Immunohistochemistry(IHC)was used to detect the expression and change of megakaryocyte platelet adhesion molecule(CD36)in the spleen.Results:There were abundant red pulp and rarely white pulp at P1 d spleen.White pulp and marginal and marginal sinuses were gradually formed at P5 d.The structure of the developed spleen in mice was gradually close to the spleen of adult mice.The megaconuclear cells were clustered in group and mostly distributed between the capsule,around blood vessels and splenic cord.Three types of megakaryocytes including granule type,platelet type and naked karyotype could be distinguished by CD36 IHC staining.There was a gradually increasing trend of granule type and naked karyotype,but a declining trend was seen in platelet type karyotype.Conclusion:The morphology of spleen is close to adult′s at P31 d which is capable of hematopoiesis for life.CD36 immunohistochemistry can divide megakaryocytes into granule type,platelet type and naked karyotype.
