摘要
目的:探索褪黑素抑制胃癌细胞作用及分子机制。方法:采用褪黑素处理人胃癌细胞系AGS和MGC803,倒置显微镜下观察细胞形态及结构变化;流式细胞仪检测褪黑素对细胞周期、细胞增殖的影响;并分析与细胞周期、细胞增殖相关蛋白CDC25A、p21表达及磷酸化水平来确定褪黑素对胃癌细胞作用的分子机制。结果:褪黑素作用后,AGS和MGC803细胞形态发生明显变化;细胞增殖相关蛋白CDC25A、p21表达及磷酸化下调;G0/G1期细胞上升,S期和G2/M期细胞下降,引起细胞周期阻滞在G1/S期,抑制胃癌细胞增殖。结论:褪黑素通过下调CDC25A、p21表达及磷酸化水平,引起AGS和MGC803胃癌细胞周期阻滞在G1/S期,抑制胃癌细胞增殖。
To explore the effect of melatonin on gastric cancer cells and its molecular mechanism.Methods:Melatonin was used to treat human gastric cancer cell lines AGS and MGC803.Cell morphology and structural changes were observed under inverted microscope.Flow cytometry was used to detect the effect of melatonin on cell cycle and cell proliferation.The expression and phosphorylation of protein CDC25A,p21 related to cell cycle and cell proliferation were analyzed to determine the molecular mechanism of melatonin on gastric cancer cells.Results:After melatonin treatment,morphological characteristics changed significantly;the expression and phosphorylation of cell proliferation related proteins CDC25A,p21 were down-regulated;G0/G1 increased,S and G2/M decreased that inferred melatonin induced cell cycle arrest at the G1/S phase and inhibited cell proliferation in AGS and MGC803 cells.Conclusion:Melatonin down-regulates the expression and phosphorylation of CDC25A and p21,which leads to cell cycle arrest at the G1/S phase thus inhibiting the cell proliferation in gastric cancer cells.
