血清与全反式维甲酸对神经干细胞分化及突起生长的作用差异 被引量:1

Comparison of the effects of serum and all-trans-retinoic acid on the differentiation of neural stem cells and the neurite outgrowth in vitro

摘要 目的:比较血清与全反式维甲酸(RA)2种诱导条件对神经干细胞分化及神经元突起生长作用的差异。方法:取E12.5d的胚胎小鼠皮层分离培养得到神经干细胞,然后分别用低浓度2%的血清和1μmol/L RA诱导神经干细胞分化3d和7d,用免疫荧光鉴定神经干细胞分化特征。结果:RA诱导组中的神经元比例显著高于血清诱导组,长期诱导至7d其仍保持较高的水平,但其星形胶质细胞比例明显低于血清诱导组;随着诱导时间的延长,RA诱导组中神经元突起的长度和数目也都显著长于和高于血清诱导组;另外,血清和RA诱导形成的胶质细胞的形态特征不一致。结论:RA相较于血清更利于诱导神经元的分化以及突起的生长,而血清更利于诱导星形胶质细胞的分化。 To compare the effects of serum and all-trans-retinoic acid(RA)on the differentiation of neural stem cells as well as the neurite outgrowth of neurons in vitro.Methods:Neural stem cells were isolated and cultured from the cortices of E12.5 embryos in vitro,and then induced by 2%serum and 1μmol/L RA respectively for 3 days and 7 days.Immunofluorescence was employed to identify the differentiation of neural stem cells.Results:The proportion of neurons in the RA induced group was significantly higher than that in the serum induced group,and it remained high after the long-term induction to 7 days,while the proportion of astrocytes was significantly lower than that in the serum induced group.With the prolongation of induction time,the length and number of neurites in RA induced group were significantly longer or higher than those in the serum induced group.In addition,the morphological characteristics of glial cells induced by serum and RA were not the same.Conclusion:RA is more conducive to induce neuronal differentiation of neural stem cells and neurite outgrowth,but serum is more beneficial to promote astrocytic differentiation of neural stem cells.
