摘要
[目的]研究穿心莲有效部位中穿心莲内酯在小鼠体内的组织分布。[方法]小鼠单次灌胃给予穿心莲内酯单体(223 mg/kg)和穿心莲有效部位(600 mg/kg),1. 5 h时取心、肝、脾、肺、肾、脑、胃、肠及肌肉组织,高效液相色谱法测定各组织中穿心莲内酯的浓度。[结果]给予穿心莲内酯单体和穿心莲有效部位1. 5 h后,穿心莲内酯在小鼠各组织中均有分布,在胃、肠中分布相对较多;给予穿心莲有效部位后,各组织(除脑外)中穿心莲内酯含量均较给予穿心莲内酯单体后高,特别是肝、肾、胃和肌肉等组织。[结论]与穿心莲内酯单体相比,小鼠对穿心莲有效部位中的穿心莲内酯吸收率更高,生物利用度更高。
〔Objective〕To study tissue distribution of andrographolide in mice after oral administration of the active ingredient of Andrographis paniculata(burm.f.)Nees.〔Methods〕Mice were administrated with androgropholide(223mg/kg)or the active ingredient of Andrographis paniculata(burm.f.)Nees(600mg/kg).1.5hours later,the organs of those mice such as(heart,liver,spleen,lung,kidney,brain,stomach,intestine and muscle)were biopsied and the concentration of andrographolide was accurately determined.〔Results〕1.5hours after the mice were administrated with androgropholide or the active ingredient of Andrographis paniculata(Burm.f.)Nees,andrographolide was detected in each tissue individually.The concentration of andrographolide was higher in some organs rather than the others such as stomach and intestine.Compared to andrographolide monomer,the concentration was higher when the mice were administrated with the active ingredient of Andrographis paniculata(Burm.f.)Nees especially in some organs such as liver,kidney,stomach and muscle.〔Conclusion〕The active ingredient of Andrographis paniculata(Burm.f.)Nees can significantly increase the absorption and bioavailability of andrographolide.
