不同药物麻醉的乳腺癌患者血清对乳腺癌MDA-MB-231细胞凋亡的影响 被引量:11

Effects of serum from breast cancer patients undergoing different drug anesthesia on apoptosis of breast cancer MDA-MB-231 cells

摘要 目的:探究不同药物麻醉的乳腺癌患者血清对体外乳腺癌MDA-MB-231细胞凋亡的影响。方法:收集我院2016年2月至2017年12月期间采用丙泊酚麻醉的乳腺癌患者血液样本28例,七氟醚麻醉的乳腺癌患者血液标本32例,乳腺癌术后镇痛使用曲马多的患者血液样本25例,未使用任何麻醉药的普通乳腺癌患者血液样本20例。将血清作用于乳腺癌MDA-MB-231细胞24 h后,采用流式细胞法检测细胞的凋亡率,ELISA法检测各组细胞中半胱氨酸蛋白酶3(caspase-3)和半胱氨酸蛋白酶8(caspase-8)的表达情况。结果:流式细胞法检测结果显示,使用丙泊酚、七氟醚麻醉和曲马多术后镇痛的患者血清均能够显著提高乳腺癌MDA-MB-231细胞凋亡率; ELISA结果显示,使用丙泊酚、七氟醚麻醉和曲马多术后镇痛的患者血清均能够显著提高乳腺癌MDA-MB-231细胞caspase-3的表达(P <0. 05),且各组之间无统计学差异(P> 0. 05);而使用丙泊酚和七氟醚麻醉的患者血清均能够显著提高乳腺癌MDA-MB-231细胞caspase-8的表达(P <0. 05),曲马多却未能影响乳腺癌MDA-MB-231细胞caspase-8的表达(P> 0. 05),丙泊酚和七氟醚组与曲马多组对比,差异具有统计学意义(P <0. 05)。结论:使用丙泊酚、七氟醚麻醉和曲马多术后镇痛的患者血清均能够显著提高乳腺癌MDA-MB-231细胞凋亡率及caspase-3的表达;而使用丙泊酚和七氟醚麻醉的患者血清能够显著提高乳腺癌MDA-MB-231细胞caspase-8的表达,曲马多对乳腺癌MDAMB-231细胞caspase-8的表达没有明显影响。 Objective:To investigate the effect of serum from patients with breast cancer with different drugs anes-thesia on the apoptosis of breast cancer MDA-MB-231cells in vitro.Methods:28blood samples from patients with breast cancer anesthetized with propofol,32patients with sevoflurane anesthetized,blood samples of25patients after tramadol analgesia after breast cancer operation and20patients with normal breast cancer without any anesthetics from February2016to December2017in our hospital were collected.After sera were treated with MDA-MB-231cells for24h,the apoptosis rate of the cells was detected by flow cytometry.Expression of caspase-3and caspase-8were detected by ELISA.Results:Flow cytometry results showed that the use of propofol,sevoflurane anesthesia and tramadol postoperative analgesia can significantly increase the apoptosis of breast cancer MDA-MB-231cells.ELISA results show that the use of different the serum levels of propofol,sevoflurane anesthesia,and tramadol postoperative analge-sia significantly increased the expression of caspase-3in breast cancer MDA-MB-231cells(P<0.05),and there was no statistical difference between the groups(P>0.05).Serum from patients anesthetized with propofol and sevoflurane significantly increased the expression of caspase-8in breast cancer MDA-MB-231cells(P<0.05),but tramadol failed to affect the expression of caspase-8in breast cancer MDA-MB-231cells(P>0.05),the difference between propofol and sevoflurane groups and tramadol group were statistically significant(P<0.05).Conclusion:Patients treated with propofol,sevoflurane anesthesia,and postoperative analgesia with tramadol significantly increased the rate of apoptosis and caspase-3expression in breast cancer MDA-MB-231cells.The serum of patients with propofol,sevoflurane anesthesia can significantly increase the expression of caspase-8in breast cancer MDA-MB-231cells.Tramadol has no significant effect on the expression of caspase-8in breast cancer cells MDA-MB-231.
