杨桃根提取物对CCl_4致小鼠急性肝损伤的预防作用 被引量:8

摘要 目的探讨杨桃根提物(EACR)对CCl_4致小鼠急性肝损伤的影响。方法预防性给予相应药物7 d后,复制急性肝损伤模型。检测血清中的谷草转氨酶(AST)、丙氨酸氨基转移酶(ALT)、白细胞介素1(IL-1)、白细胞介素6(IL-6)的水平,以及肝脏组织中超氧化物歧化酶(SOD)、丙二醛(MDA)、还原型谷胱甘肽(GSH)、谷胱甘肽过氧化物酶(GSH-Px)的活性;Western blot检测肝脏组织中肿瘤坏死因子α(TNF-α)、核转录因子-κB(NF-κB)、caspase-3的蛋白表达水平;HE染色观察小鼠肝脏的病变情况。结果与模型组相比,给药组小鼠血清中AST、ALT、IL-1、IL-6及肝脏中MDA水平明显降低;肝脏组织中SOD、GSH、GSH-Px明显升高;肝脏组织TNF-α、NF-κB、caspase-3蛋白表达明显减少;HE染色显示肝脏组织病变减轻。结论 EACR通过降低酶活性,抑制脂质过氧化及炎症因子释放,达到保护小鼠肝脏的作用。 Aim To investigate the effect of extract of Averrhoa carambola L.root(EACR)on carbon tetrachloride(CCl4)induced acute hepatic injury in mice.Methods Both NS and the treatment protocol were administered via intragastric gavage(i.g.)for seven days.Each group of mice were intraperitineally injected with0.15%CCl4to establish an acute liver injury model except for the normal group.The levels of aspartate aminotransferase(AST),alanine aminotransferase(ALT),interleukin-1(IL-1),interleukin-6(IL-6)in the serum and the superoxide dismutase(SOD),malondialehyde(MDA),glutathione(GSH),and glutathione peroxidase(GSH-Px)in the liver tissues were measured.The protein expression of tumor necrosis factor-α(TNF-α),nuclear factor-kappa B(NF-κB),caspase-3were measured by western blot.The histopathological changes were detected by HE staining.Results After pretreatment with EACR,the levels of AST,ALT,IL-1,IL-6in serum as well as MDA in the liver markedly decreased,whereas the activities of SOD,GSH and GSH-Px increased.The protein expressions of TNF-α,NF-κB and caspase-3were significantly down-regulated in EACR groups.HE staining also showed that the injury of liver was mitigated after administration of EACR.Conclusions EACR can prevent CCl4induced acute hepatic injury in mice.The mechanism may be related to attenuating the free radicals and inhibiting the lipid peroxidation.
