基于斑马鱼模型对α-倒捻子素抗血管生成活性的初步探讨 被引量:1

摘要 目的:研究α-倒捻子素的抗血管生成活性。方法:首先我们测定了α-倒捻子素对斑马鱼的急性毒性,然后在安全浓度条件下,采用转基因斑马鱼和野生型斑马鱼模型分别通过不同的检测方法来测定α-倒捻子素的抗血管生成活性。结果:α-倒捻子素能够抑制转基因斑马鱼节间血管的生成数目,并且呈现剂量依赖性;α-倒捻子素能够降低72 hpf野生型斑马鱼胚胎体内的碱性磷酸酶含量,呈现剂量相关性。结论:α-倒捻子素具有明显的抗血管生成活性。 Objective:To explore the potent anti-angiogenic effect of α-Mangostin in zebrafish model .Methods:First,α-Mangostin was determined for acute toxicity to zebrafish ,then, The anti-angiogenic activities of theα-Mangostin was evaluated in the zebrafish model by two different methods in transgenic zebrafish and wild zebrafish.Results:The result showed that α-Mangostin can reduce the number of intersegmental blood vessel formed and reduce the level of quantitative endogenous alkaline phosphatase in transgenic zebrafish and wild zebrafish, respectively, which displayed strong anti-angiogenic activity in a way of dose-related.Conclusion:α-Mangostin is a potent anti-angiogenic component and can serve as a potent inhibitor in the angiogenesis in the zebrafish embryo model.
