摘要
本文报道一种简洁而有效的方法来合成Lifitegrast中间体苯并呋喃-6-甲酸。该反应以廉价易得的3-羟基-4-碘苯甲酸为原料,通过Sonogashira偶联反应、碘化亚铜催化关环、碱性水解得到目标化合物苯并呋喃-6-甲酸,三步反应总收率62%。中间体及产物结构经1H NMR、13C NMR和ESI-MS确证。
Benzofuran-6-carboxylic acid,the key intermediate of Lifitegrast,was synthesized by an efficient and parctical method from methyl 3-hydroxy-4-iodobenzoate througha three-step procedurein a total yield of 62%,involving Sonogashira coupling reac-tion,cyclization in the presence of cuprous iodide,hydrolysis of ester.The structures of intermediates and the product were con-firmed by 1 H NMR,13 C NMR and ESI-MS.
