摘要
目的探讨CYP1A1在孕期暴露香烟烟雾所致宫内发育迟缓(IUGR)胎鼠胎盘组织中的表达及其意义。方法在自制熏烟箱中制备IUGR模型,比较模型组和正常对照组胎盘中的药物代谢酶CYP1A1及细胞凋亡相关蛋白的表达量差异。结果 IUGR模型组胎鼠的平均体重、身长、胎盘重量均明显低于对照组(P均<0. 001)。免疫组织化学结果显示,与对照组相比,模型组胎盘中CYP1A1表达量升高,Bcl2蛋白在胎盘中的表达降低(P <0. 001),BAX蛋白表达增高(P <0. 001)。结论孕期吸烟可致金黄地鼠胎盘药物代谢酶表达升高,提示香烟烟雾中有害物质可以通过母体到达胎盘,引起胎盘细胞的死亡,影响胎盘的发育,最终导致IUGR。
Objective To investigate the expression and significance of CYP1A1in the placental tissue of fetal rats with intrauterine growth retardation(IUGR)induced by passive cigarette smoking during pregnancy.Methods The model of fetal IUGR was established in a homemade smoker box.We compared the expression levels of the drug-metabolizing enzymes CYP1A1and apoptosis-related proteins in the placenta of the model group and the normal control group.Results The average body weight,body length and placental weight of fetus in the IUGR model group were significantly lower than those in the control group(P<0.001).The immunohistochemical staining results showed that the expression of CYP1A1in the placenta of the model group was higher than that in the control group.Compared with the control group,the expression of Bcl-2in the placenta of the model group was decreased(P<0.001),while the BAX was increased(P<0.001).Conclusion Smoking during pregnancy could cause the overexpression of drug-metabolizing enzymes in the fetal placenta of golden hamsters,suggesting that the harmful substances of cigarette smoke could be transferred to fetus via placenta,which could lead to the death of placental cells,affect the development of placenta,and ultimately result in IUGR.
