摘要
以黄肉品种桃果实‘金丽’为试验材料,利用同源序列克隆技术获得桃果实番茄红素β-环化酶(LCYb)基因的全长核苷酸序列,命名为PpLCYb。PpLCYb全长1 699bp,编码区长度1 515bp,编码504个氨基酸。进化树分析发现PpLCYb与草莓FaLCYb亲缘性较高。氨基酸序列分析表明PpLCYb含有特征序列LYAMPF及NAD(P)/FAD结合区,属于SDR超级家族。实时荧光定量PCR结果显示PpLCYb基因在桃果实中的表达总体呈现上升的趋势,并在果实处于硬熟期时达到较大值,随后在完熟期略有下降。桃果实PpLCYb基因的克隆及表达分析可为研究桃果实类胡萝卜素生物合成分子机制奠定良好的基础。
Using homologous sequence cloning method,full-length cDNAs of lycopeneβ-cyclase(LCYb)was isolated from‘Jinli’peach fruit,a yellow peach variety,and named as PpLCYb.PpLCYb was1699bp in full length and encoded a predicted protein of504amino acids(ORF length1515bp).Phylogenetic analysis indicated that PpLCYb was similar to FaLCYb from Fragariax ananassa.Deduced amino acid sequence demonstrated that PpLCYb contained characteristic sequence LYAMPF and NAD(P)/FAD binding domain,which belonged to the SDR super family.The quantitative real-time PCR showed that PpLCYb transcripts showed an upward tendency and reached a larger value during fruit hard-maturing period and then decreased slightly in maturity.For the cloning and expression analysis of PpLCYb from peach fruit was reported,it lays the foundation for further research of carotenoid biosynthetic pathway.
