摘要
为花青素提取开辟新途径并实现山竹果皮废物资源化利用,以山竹果皮为原料,采用单因素结合正交试验研究提取时间、温度、料液比及乙醇浓度对山竹果皮中花青素提取效果的影响;采用AB-8大孔树脂纯化花青素提取物,并检测其清除2-联氮-二(3-乙基-苯并噻唑-6-磺酸)二铵盐自由基(ABTS+·)和1,1-二苯基-2-苦肼基(DPPH·)自由基的能力。结果表明:山竹果皮提取花青素的最佳条件为提取温度75℃,料液比1∶30,水浴提取11h,乙醇浓度60%,此条件下花青素提取量为2 083.5nmol/g。AB-8大孔树脂吸附2h后在pH 1.0的70%乙醇溶液解吸2h,吸附率为32.9%,解吸率为74.3%。此纯化物对2种自由基均具有清除能力,0.024mg纯化物对ABTS+·的清除率为13.8%,对DPPH的清除率为44.3%。
The effect of extraction time,temperature,solid/liquid ratio and ethanol concentration on extraction of anthocyanin from Garcinia mangostana peel was studied by the single-factor and orthogonal tests and the scavenging ABTS+·and DPPH·capacity of the anthocyanin extract purified with AB-8 macroporous resin was detected to explore new pathways for extraction of anthocyanin and realize wastes resources utilization of G.mangostana peel.Result:The optimum extraction conditions for anthocyanin extraction from G.mangostana peel include 1∶30 solid/liquid ratio,60% ethanol and at 75℃ for 11 h.The anthocyanin extraction amount can reach 2 083.5 nmol/g under the optimum extraction conditions.The adsorption rate and desorption rate of the anthocyanin extract purified with AB-8 macroporous resin for 2 h and then 2 h desorption in 70%ethanol with pH 1.0 is 32.9% and 74.3% respectively and the purified anthocyanin extract is of radical-scavenging capacity to ABTS+·and DPPH·.The ABTS+·and DPPH·scavenging capacity of 0.024 mg purified anthocyanin extract is 13.8%and 44.3% respectively.
