卵类粘蛋白手性固定相拆分异丙嗪对映异构体

摘要 建立了异丙嗪对映异构体的HPLC手性拆分方法,利用卵类粘蛋白作为手性固定相,考查了流动相中有机改性剂的含量、缓冲盐溶液的浓度、流动相pH值、流速等色谱条件对手性拆分的影响.优化条件流动相为甲醇-磷酸二氢钾(0.01 mol/L)为6∶94(V/V),p H=2.96,柱温为25℃,流速为0.7 m L/min,15 min内完成异丙嗪对映异构体的手性拆分,分离度Rs=1.417.得出卵类粘蛋白手性固定相能够较好地拆分异丙嗪对映异构体. To establish the chiral separation method of promethazine.Promethazine enantiomers were separated on ovomucoid column by HPLC,the influence factors on chiral separation,including the concentration of organic modifiers,the concentration of buffer solution,the flow rate and the pH of mobile phase were considered.The results showed that the optimum mobile phase was methanol-0.01mol/L KH2PO4(6∶94),pH=2.96,the column temperature was 25℃,the flow rate was 0.7 mL/min,promethazine enantiomers were separated in 15 min (Rs=1.417).Promethazine enantiomers can be separated by HPLC with ovomucoid as chiral stationary phase.
