摘要
将细菌基因组随机打断,是构建随机文库的第一步,也是构建随机、多样文库的一个重要保障。物理法(超声波)和酶切法是两种常用的断裂基因组的方法。本试验采用超声法和酶切法对马耳他布鲁菌16M株基因组进行随机打断,并比较二者的打断效果,为后续建立布鲁菌基因组随机文库奠定基础。结果表明,用超声法(超声条件:输出功率50%,超声3 s,间隔3 s,超生次数10下)随机打断布鲁菌基因组效果最好。
The interruption of genome is the first step in constructing libraries.And it is also an important guarantee to build random and diverse library.Physical methods(ultrasound)and enzyme digestion are common methods for breaking genomes.In this experiment the genome of Brucella melitensis 16M strain was randomly interrupted by ultrasonic and enzyme digestion.And the effect of the two methods was compared to lay the foundation for the subsequent establishment of random library.The results showed that the genome was best randomly interrupted by ultrasonic under the condition that output power of 50%,ultrasonic 3S,interval 3S and 10 excess times.
