摘要
目的观察膀胱癌细胞及组织中长链非编码RNA(LncRNAs)尿路上皮癌抗原1(UCA1)和miR-195的表达及其与膀胱癌患者临床病理特征的关系,探讨UCA1和miR-195与膀胱癌患者预后的关系。方法选取人膀胱上皮永生化细胞株SV-HUC-1作为对照组,膀胱癌细胞株T24为膀胱癌细胞组。选择经根治性膀胱切除术后患者的膀胱上皮癌组织及癌旁正常组织各50例,收集患者的病理分级、临床分期、肿瘤浸润、复发等资料。采用RTPCR法检测膀胱癌细胞及组织中UCA1、miR-195表达,采用Pearson相关性分析法分析UCA1与miR-195表达的相关性。术后对50例患者随访3年,采用Cox回归模型分析影响患者预后的因素。结果膀胱癌细胞组UCA1表达高于对照组、miR-195表达低于对照组(P均<0. 05)。膀胱癌组织中UCA1表达高于癌旁正常组织、miR-195表达低于癌旁正常组织(P均<0. 05)。病理分级较高、临床分期T3~T4者、肿瘤浸润及复发者UCA1表达较高,病理分级低者、临床分期T1~T2者、肿瘤未浸润及初发者miR-195表达较高(P均<0. 05)。Pearson相关性分析示,膀胱癌组织中UCA1与miR-195表达呈负相关(r=-0. 673,P <0. 05)。临床分期T1~T2患者生存率高于T3~T4期患者,UCA1阴性表达患者生存率高于阳性表达患者,miR-195阳性表达患者生存率高于阴性表达患者(P均<0. 05)。Cox回归模型示UCA1阳性、miR-195阴性、临床分期T3~T4及肿瘤转移是影响膀胱癌患者预后的独立危险因素(P均<0. 05)。结论膀胱癌细胞及组织中LncRNA UCA1表达增高、miR-195表达降低,UCA1表达增高、miR-195表达降低均与膀胱癌患者的预后不良显著相关。
Objective To investigate the expression levels of urothelial cancer associated1(UCA1)and miR-195in the bladder cancer cells and tissues,its correlation with the clinicopathologic features of patients with bladder cancer,and its role in the progress of bladder cancer.Methods The human bladder epithelial immortalized cell line(SV-HUC-1)was selected as the control group,and bladder cancer cell line(T24)was selected as bladder cancer cell group.After radical cystectomy,50cases of bladder epithelial carcinoma tissues and50cases of adjacent normal tissues were collected.The data of pathological grade,clinical stage,tumor invasion and recurrence were collected.We detected the mRNA expression of UCA1and miR-195in the bladder cancer cells and bladder epithelial carcinoma tissues by RT-PCR.All patients were followed up for3years.Cox regression model was used to analyze the prognostic factors,and Pearson correlation analysis was used to analyze the correlation between mRNA expression of UCA1and miR-195.Results The expression of UCA1mRNA in the bladder cancer cells was higher than that in the control group,and the expression of miR-195mRNA was lower than that in the control group(both P<0.05).The expression of UCAl mRNA in the bladder cancer tissues was higher than that in the adjacent normal tissues,and the expression of miR-195mRNA was lower than that in the adjacent normal tissues(both P<0.05).The expression of UCA1was higher in patients with the clinical features of higher pathological grade,clinical stage T3-T4,tumor invasion and recurrence,and the expression of miR-195was higher in patients with the clinical features of low grade,clinical stage T1-T2,uninfiltrated tumor and primary tumor(both P<0.05).The survival rate of patients in clinical stage T1-T2was higher than that of patients in T3-T4stage,the survival rate of patients with UCA1negative expression was higher than that of patients with UCA1positive expression,and the survival rate of patients with miR-195negative expression was lower than that of patients with miR-195positive expression(all P<0.05).Cox regression model showed that UCA1positive expression,miR-195negative expression,clinical stage T3-T4and tumor metastasis were independent risk factors for the prognosis of bladder cancer patients(all P<0.05).Pearson correlation analysis showed that there was a negative correlation between UCA1and miR-195mRNA expression in the bladder epithelial carcinoma tissues(r=-0.673,P<0.05).Conclusion In the bladder cancer cells and tissues,the mRNA expression of UCAl is increased,and mRNA expression of miR-195is decreased,and meanwhile,the high expression of UCAl and low expression of miR-195are associated with the tumor metastasis and poor prognosis of bladder cancer significantly.
