黄芪赤风汤对大鼠胶原诱导性关节炎的治疗作用

Therapeutic effect of Huangqichifeng decoction on collagen-induced arthritis in rats

目的探讨黄芪赤风汤对大鼠胶原诱导性关节炎的治疗作用。方法选择雄性SD大鼠60只,随机分为对照组、模型组、甲氨蝶呤(MTX)组及黄芪赤风汤低、中、高剂量组,每组10只。模型组、MTX组及黄芪赤风汤低、中、高剂量组建立胶原诱导性关节炎模型,对照组不建模,正常喂饲。建模次日,MTX组给予MTX 0. 3 mg/kg灌胃,黄芪赤风汤低、中、高剂量组分别给予含黄芪赤风汤生药量7. 7、23. 1、69. 3 g/kg灌胃,对照组和模型组给予等体积蒸馏水灌胃,连续灌胃21天。模型组、MTX组及黄芪赤风汤低、中、高剂量组分别于建模第13、16、19、21天,按5级评分法进行双后踝关节AI评分;建模第0、7、13、16、19、21天,用游标卡尺测量双后踝关节直径。对照组同时间进行AI评分并测量双后踝关节直径。末次灌胃24 h,腹主动脉取血,采用ELISA法检测血清TNF-α、IL-1β、IFN-γ;取双后膝关节滑液,采用ELISA法检测膝关节滑液瓜氨酸化组蛋白H3。结果模型组AI评分、双后踝关节直径和血清TNF-α、IL-1β、IFN-γ含量及关节滑液瓜氨酸化组蛋白H3含量均较对照组明显增加(P均<0. 05)。与模型组比较,黄芪赤风汤低剂量组和MTX组AI评分、踝关节肿胀均有一定程度改善,且其血清TNF-α、IL-1β、IFN-γ含量及关节滑液瓜氨酸化组蛋白H3含量均明显降低(P均<0. 05)。结论黄芪赤风汤可用于治疗大鼠胶原诱导性关节炎。

Objective To explore the therapeutic effect of Huangqichifeng decoction on collagen-induced arthritis in rats.Methods Sixty male SD rats were randomly divided into the blank control group,model group,methotrexate(MTX)group,low-dose,medium-dose and high-dose Huangqichifeng decoction groups,with10rats in each group.In addition to the blank control group,we established the collagen-induced arthritis models in the other groups,and each group was fed normally.On the next day of modeling,MTX group was given 0.3mg/kg MTX intragastrically(twice a week for three weeks),and low-dose,medium-dose and high-dose groups were given7.7,23.1,and69.3g/kg of crude drug containing Huangqichifeng decoction.The control group and the model group were given an equal volume of distilled water for intragastric administration,and all groups were continuously administered for21days except the MTX group.The arthritis index of the double posterior ankle joints of each group were scored according to the5-level scale on the13th,16th,19th and21st days of modeling,respectively.The diameter of the double posterior ankle joint was measured with a vernier caliper,on day0,7,13,16,19,and21of modeling.At24h after the last gavage,the blood was taken from the abdominal aorta and the bilateral posterior knee synovial fluid was taken out.The levels of TNF-α,IL-1β,IFN-γin serum and citrullinated histone H3in synovial fluid were determined by ELISA.Results The AI,bilateral posterior ankle diameter,TNF-α,IL-1β,IFN-γ content in serum and citrullinated histone H3 content in synovial fluid of the model group increased significantly as compared with those of the control group(all P<0.05).Compared with the model group,the AI and ankle swelling in the low-dose group and the MTX group were improved to some extent,the content of TNF-α,IL-1β,IFN-γ and citrullinated histone H3 were reduced significantly(P<0.05).Conclusion Huangqichifeng decoction can be used to treat collagen-induced arthritis in rats.
