肠炎型犬细小病毒的分离鉴定及病毒滴度测定 被引量:4

摘要 为获得肠炎型犬细小病毒分离株,采集犬细小病毒胶体金初检阳性的3份粪便,采用同步接毒法分别接种猫肾细胞(F81)和犬肾细胞(MDCK)进行分离培养,F81盲传至第2代出现细胞病变,MDCK盲传至第3代出现细胞病变,2种细胞均仅从病料3中分离出1株病毒。采用PCR方法鉴定该分离株为犬细小病毒并命名为CPV3。根据VP2基因核苷酸序列绘制的系统进化树及其推导氨基酸序列的分析,确定CPV3属于CPV-2a亚型。测定CPV3在MDCK中72 h时的组织细胞半数感染量(TCID50),第5代次(CPV-M-5)和第10代次(CPV-M-10)的TCID50数值分别为10-4. 83/0. 1 mL和10-5. 50/0. 1 mL,这为后续测定重组犬干扰素活性试验提供了材料。
