氟尿嘧啶联合TNF-α对人胃癌细胞SGC-7901的抑制作用 被引量:2

摘要 目的研究5-氟尿嘧啶(5-Fu)联合肿瘤坏死因子α(TNF-α)对人胃癌细胞SGC-7901的增殖抑制及凋亡诱导的作用,并探讨其机制。方法应用噻唑蓝(MTT)比色法检测单用不同浓度5-Fu、TNF-α及两者联合应用对SGC-7901细胞增殖的抑制率;采用Hoechst检测SGC-7901细胞凋亡情况。结果不同浓度的5-Fu(6.25、12.50、25.00、50.00mg/L)均可抑制SGC-7901细胞增殖(F=32.41,P<0.05);不同浓度的TNF-α(25.00、50.00、100.00、150.00mg/L)均可抑制SGC-7901细胞增殖(F=28.92,P<0.05);5-Fu (6.25 mg/L)与TNF-α(25.00、50.00、100.00、150.00mg/L)联合应用对SGC-7901细胞的增殖抑制率较单用TNF-α或5-Fu增加(F=28.05,P<0.05)。Hoechst检测结果显示,100.00mg/L TNF-α组、6.25mg/L 5-Fu组及二者联合应用组均有细胞凋亡现象(F=124.03、46.23,P<0.05),且联合用药组细胞凋亡率增高(F=124.03,P<0.05)。结论 TNF-α可增强5-Fu对人胃癌细胞SGC-7901增殖抑制及凋亡诱导的作用,其机制可能为5-Fu、TNF-α分别激活不同的信号通路,并级联和放大相关的凋亡信号,协同抑制细胞生长,诱导细胞凋亡。 Objective To investigate the effect of 5-fluorouracil(5-Fu)combined with tumor necrosis factor-alpha(TNF-α)in the proliferation inhibition and apoptosis induction of human gastric cancer cell line SGC-7901 and its mechanism.[WTHX]Methods After the treatment with different concentrations of 5-Fu,TNF-α,or 5-Fu combined with TNF-α,the growth inhibition of SGC-7901cells was measured by MTT assay.Hoechst assay was used to assess the SGC-7901cell apoptosis.Results 5-Fu at different concentrations(6.25,12.50,25.00,and50.00mg/L)inhibited the proliferation of SGC-7901cells(F=32.41,P<0.05);TNF-α at different concentrations(25.00,50.00,100.00,and150.00mg/L)also inhibited the proliferation of SGC-7901cells(F=28.92,P<0.05);5-Fu(6.25mg/L)combined with TNF-α(25.00,50.00,100.00,and150.00mg/L)significantly increased the inhibition rate of SGC-7901 proliferation compared with TNF-α or 5-Fu alone(F=28.05,P<0.05).The Results of Hoechst assay showed that apoptosis was observed in the 100.00mg/L TNF-αgroup(F=124.03,P<0.05),the 6.25mg/L 5-Fu group(F=46.23,P<0.05),and the combination groups,and the apoptosis rate was significantly increased in the combination groups(F=124.03,P<0.05).ConclusionTNF-αcan enhance the effect of 5-Fu in the proliferation inhibition and apoptosis induction of human gastric cancer cell line SGC-7901.The mechanism may be that5-Fu and TNF-αactivate different signaling pathways,and cascade and amplify the related apoptosis signals to inhibit cell growth and induce cell apoptosis in a synergistic manner.
