摘要
溶藻弧菌是我国海水产养殖鱼类的重要病原菌,已给海水养殖业带来巨大的经济损失.利用生物信息学手段,筛选获得来自溶藻弧菌的一个sRNA分子,RyhB,并对其进行了克隆和无标记框内突变株的构建,分析了其功能.结果表明:溶藻弧菌RyhB编码基因大小为233bp,具有典型的sRNA茎环结构,同时在其-10至-35区具有Fur蛋白结合序列.RyhB在溶藻弧菌体内具有多个潜在的靶标mRNA分子,可能参与其多种生理进程.同时,ΔryhB缺失株的运动性几乎完全丧失,铁载体合成能力降低.进一步的研究丰富了人们对溶藻弧菌sRNA分子及其毒力调控机制的认识.
Vibrio alginolyticus is one of the important fish pathogen in China,bringing about serious economic loss to marine culture.In this paper,a sRNA,RyhB,was screened with the bioinformatics methods.The gene encoded for RyhB was cloned and the in-frame deletion mutant strain was constructed in Vibrio alginolyticus.Also the functions of RyhB in in Vibrio alginolyticus were characterized.The results showed that the gene encoded for RyhB is 233bp in size,demonstrated the classical stem-loop structure and contained the Fur binding sites in-10 to-35 region.There are multiple potential target mRNAs of RyhB,indicating that RyhB may be involved in many physiological processes in Vibrio alginolyticus.Besides,the ability of mobility and the siderophore production were both impaired in theΔryhB mutant strain,contrast to the wild-type strain.This paper will enrich the peoples recognition of the sRNA and virulence regulation mechanism in Vibrio alginolyticus.
