针刺肾俞足三里干预绝经后骨质疏松症大鼠模型的影响 被引量:7

The Effect of Acupuncture at Shenshu(BL23) and Zusanli(ST36) in the Treatment of Postmenopausal Osteoporosis Rat Model

摘要 目的探讨针刺肾俞足三里防治绝经后骨质疏松症(PMOP)的可能作用机制。方法 50只雌性SD大鼠随机分为空白组、假手术组、模型组、肾俞足三里组和非经非穴组。行双侧卵巢切除术复制骨质疏松模型。肾俞足三里组、非经非穴组每天针刺1次,留针30 min,10次1疗程,疗程间期5天,6个疗程后同时处死5组大鼠,取右侧胫骨测量胫骨、胫骨上端骨密度;RTPCR法检测大鼠腰椎c-myc、Cyclin D1和R unx2 mR NA表达。结果与假手术组比较,模型组胫骨、胫骨上端骨密度降低(P<0.05);与模型组比较,肾俞足三里组胫骨、胫骨上端骨密度升高(P<0.05),非经非穴组胫骨、胫骨上端骨密度差异均无统计学意义(P>0.05);c-myc mRNA表达:肾俞足三里组c-myc mRNA表达与假手术组比较差异无统计学意义(P>0.05);各实验组Cyclin D1灰度值均值差异均无统计学意义(P>0.05);Runx2 mRNA表达:与空白组比较,模型组Runx2 mRNA表达降低(P<0.05),与模型组比较,肾俞足三里组与非经非穴组的表达升高(P<0.05)。结论针刺肾俞、足三里穴可改善绝经后骨质疏松的骨密度降低,其作用机制可能与调节腰椎c-myc、Cyclin D1和Runx2 mRNA表达有关。 Objective To explore the possible mechanisms of acupuncture at Shenshu(BL23)and Zusanli(ST36)for the treatment of postmenopausal osteoporosis(PMOP).Methods Fifty female SD rats were randomly divided into a blank group,a shamoperation group,a model group,a acupoint(BL 23 and ST36)group and a non-acupoint group.The osteoporotic rat model was established by bilateral ovariectomy.The acupoint group and non-acupoint group were given acupuncture once per day.10 treatments made one session.There were 5 days at the interval between two sessions.All rats were killed and the bone mineral density(BMD)of the right tibias and proximal tibias were obtained after 6 sessions.Meanwhile,the expression of cmyc,Cyclin D1,and Runx2 mRNA in lumbar vertebra were detected by RT-PCR.Results BMD of the right tibias and proximal tibias in the model group were lower than those in the sham-operation group(P<0.05).Compared with the model group,the BMD of the right tibias and proximal tibias in the acupoint group were obviously increased(P<0.05),but no significant difference in the non-acupoint group(P>0.05).Compared with the sham-operation group,the expression of cmyc mRNA of the acupoint group had no significant differences(P>0.05).The difference in the mean grey values of Cyclin D1 in each experimental group was not statistically significant(P>0.05).The expression of Runx2 mRNA in the model group were lower than those in the blank group(P<0.05).Compared with the model group,the expression of Runx2 mRNA in the acupoint group and non-acupoint group were obviously increased(P<0.05).Conclusion Acupuncture at Shenshu(BL23)and Zusanli(ST36)could reduce PMOP and increase BMD.This action is probably associated with the regulation of the expression of cmyc,Cyclin D1,and Runx2 mRNA in lumbar vertebra.
