肾衰方及其拆方对肾间质纤维化Wnt4、β-catenin蛋白表达的影响

Effects of shen shuai fang and its disassembly formula on the expression of renal interstitial fibrosis Wnt4 and β-catenin

目的观察中药肾衰方及其拆方对大鼠肾间质纤维化肾组织Wnt4、β-catenin蛋白表达的影响。方法将72只大鼠随机分为:假手术组、模型组、肾衰方组、祛邪方组、补虚方组、贝那普利组,采用单侧输尿管梗阻动物模型,术后7、14 d分别观察检测24 h尿蛋白定量、血清肌酐、尿素氮水平以及梗阻侧大鼠肾组织Wnt4、β-catenin含量的表达。结果 7、14 d时模型组大鼠24 h尿蛋白定量、血尿素氮、血清肌酐、Wnt4、β-catenin水平均高于假手术组(P <0.05),与模型组比较,经肾衰方、祛邪方、补虚方和贝那普利治疗后24 h尿蛋白定量、血尿素氮、血清肌酐、Wnt4、β-catenin水平明显降低(P <0.05),第14 d肾衰方优于其拆方(祛邪方、补虚方)及贝那普利(P <0.05)。结论肾衰方及其拆方可能是通过抑制Wnt4、β-catenin蛋白的表达,减轻肾间质的纤维化,改善肾功能,从而延缓慢性肾脏病进展,且肾间质纤维化中医为本虚标实之证。

Objective To observe the effect of shen shuai fang and its disassembly formula on the expression of Wnt4 and β-catenin in rat renal interstitial fibrosis. To observe the effect of shen shuai fang and its compo-nents on the expression of Wnt4/β-catenin in in rat renal interstitial fibrosis and its anti-renal interstitial fibrosis mechanism.Methods 72 rats were randomly divided into:control group,model group,shen shuai fang,elimi-nate pathogenic,tonify deficiency group,and captopril group. The animal model of unilateral ureteral obstruction(UUO)in 7 days,14 days after feeding respectively were generated to observe 24 h urine protein quantitative detection,serum creatinine and urea nitrogen levels and obstruction rat renal kidney Wnt4,β-cateninthe expres-sion of and content.Results 7 days,14 days,the model group rats 24 h urine protein,blood urea nitrogen,serum creatinine and quantitative Wnt4,β-catenin levels were higher than those in control group(P<0.05). Compared with model group,the excess or insufficiency of kidney failure,evil spirits,and that captopril treatment after 24 h urine protein,blood urea nitrogen,serum creatinine and quantitative Wnt4,β-catenin level significantly decreased(P < 0.05),and shen shuai fang was superior to eliminate pathogenic party,party and that,captopril group(P < 0.05) for 14 days.Conclusions The decoction of shen shuai fang and its components may reduce the fibrosis of renal interstitial substances,improve renal function,and delay the progression of chronic renal disease by inhibiting the expression of Wnt4 and β-catenin. In addition,the pathogenesis of renal interstitial fibrosis in traditional Chinese medicine is asthenia in origin and asthenia in superficiality.
