黄芪甲苷Ⅳ对非酒精性脂肪肝小鼠肝脏脂质代谢的作用 被引量:11

摘要 目的探讨黄芪甲苷Ⅳ(AS-Ⅳ)对非酒精性脂肪肝(NAFLD)小鼠肝脏脂质代谢的影响。方法 5周龄雄性C57BL/6小鼠随机分为正常饮食组、高脂高糖组、高脂高糖+AS-Ⅳ[30 mg/(kg·d)]组、高脂高糖+AS-Ⅳ[60 mg/(kg·d)]组、高脂高糖+AS-Ⅳ[120 mg/(kg·d)]组。高脂高糖饮食构建NAFLD小鼠动物模型,同时用不同浓度的AS-Ⅳ[30、60、120 mg/(kg·d)]进行灌胃。正常饮食组给予等体积1%羧甲基纤维素(1%CMC)灌胃,每周测体质量、随机血糖,干预13周后,处死小鼠,留取并称量肝脏,采用H&E染色以及油红O染色检测小鼠肝脏脂质沉积情况。结果高脂高糖诱导的NAFLD小鼠较正常饮食组小鼠的体质量、肝脏湿重及随机血糖增加(P <0.05);AS-Ⅳ未显著减轻NAFLD小鼠的体质量、肝脏湿重及随机血糖(P> 0.05)。与正常饮食组相比,高脂高糖诱导的NAFLD小鼠肝脏脂滴形成增加(P <0.05);而与高脂高糖组相比,不同浓度的AS-Ⅳ干预后,小鼠肝脏脂滴形成减少,以浓度为60 mg/(kg·d)的AS-Ⅳ组最明显(P <0.01)。结论黄芪甲苷Ⅳ能改善非酒精性脂肪肝小鼠的肝脏脂质沉积。 Objective To investigate the effect of astragaloside Ⅳ on liver lipid metabolism of nonalcoholic fatty liver disease in mice.Methods C57 BL/6 mice,five-week-old,male,were randomly divided into 5 groups,the normal diet group(n = 8),high-fat and high-sugar diet(HFD)group(n = 8),HFD+AS-Ⅳ[30 mg/(kg·d)]group(n = 8),HFD+AS-Ⅳ[60 mg/(kg·d)](n = 8)and HFD+AS-Ⅳ[120 mg/(kg·d)]group(n = 8). A high-fat and high-sugar diet was used to make an animal model of NAFLD mice,and gavage was performed with different concentra-tions of AS-Ⅳ[30,60,120 mg/(kg·d)]. The normal diet group was given an equal volume of 1% carboxymethylcel-lulose(1% CMC). Body weight and random blood glucose were measured weekly. After 13-week intervention,mice were anesthetized with 1% pentobarbital sodium and were sacrificed by cervical dislocation. The livers were collected and weighed,and liver lipid deposition was detected by H&E staining and oil red O staining,respectively.Results Body weight,liver weight and random blood glucose of NAFLD mice induced by high-fat and high-sugar diet were higher than those in the normal diet group(P < 0.05,respectively). AS-Ⅳ did not significantly reduce body weight,liver weight and random blood glucose in NAFLD mice(P > 0.05). Compared with the normal diet group,the liver lipid droplet formation was increased in NAFLD mice induced by high-fat and high-sugar diet(P < 0.05). Compared with the high-fat and high-sugar diet group,liver lipid droplet formation was decreased after intervention of astragaloside Ⅳ[30,60,120 mg/(kg·d)],especially in the AS-Ⅳ group with the concentration of 60 mg/(kg·d)(P <0.01).Conclusion Astragaloside Ⅳ improves liver lipid deposition in NAFLD mice.
