摘要
目的探究乳腺癌易感基因1-mRNA(BRCA1-mRNA)水平及启动子区甲基化状态与乳腺癌患者临床病理特征的相关性。方法选取2016年2月至2018年2月于西安交大一附院确诊为乳腺癌的350例患者为实验组,另选取同期行体检的50例健康女性为对照组,分别检测两组受检者的BRCA1-mRNA水平、启动子区甲基化状态,分析BRCA1-mRNA水平与启动子区甲基化状态相关性。结果实验组BRCA1-mRNA水平低于对照组,启动子区甲基化阳性状态比率显著高于对照组(P <0.05);实验组患者BRCA1-mRNA水平、启动子区甲基化状态与其年龄、TNM分期无相关性,前者与肿瘤大小存在相关性(P <0.05);后者与组织学分级存在相关性(P <0.05)。结论乳腺癌患者BRCA1-mRNA水平与肿瘤大小有关,启动子区甲基化状态与其组织学分级有关,可以作为判断乳腺癌肿瘤状态的实验室指标。
Objective To investigate the correlation between breast cancer susceptibility gene BRCA1-mRNAs,promoter methylation status and clinicopathological features of breast cancer patients.Methods From February 2016 to February 2018,350 patients with breast cancer diagnosed in the first affiliated Hospital of Xian Jiaotong University were selected as the experimental group,and 50 healthy women who underwent physical exami-nation were selected as the control group. The BRCA1-mRNA levels of the two groups were measured respectively.The methylation status of promoter region was analyzed and the correlation between BRCA1-mRNA level and pro-moter methylation state was analyzed.Results The level of BRCA1-mRNA in the experimental group was lower than that in the control group,and the positive rate of methylation in the promoter region was significantly higher than that in the control group(P < 0.05),but there was no correlation between the level of BRCA1-mRNA and the methylation status of the promoter region in the experimental group. The former was correlated with tumor size(P <0.05)and the latter with histological grade(P < 0.05).Conclusion The level of BRCA1-mRNA in breast cancer patients is related to the tumor size,and the methylation status of promoter region is related to the histological grade,which can be used as a laboratory index to judge the tumor status of breast cancer.
