摘要
目的研究miR-9和miR-34c在老年痴呆患者血清中的表达水平,分析其对老年痴呆的诊断价值。方法以本院接收诊治的73例老年痴呆患者为研究对象,同时选取70例将健康体检者作为正常组,采用RTq PCR检测所有研究者血清中miR-9和miR-34c的水平,采用临床痴呆评定量表(CDR)对老年痴呆患者进行认知能力评分,观察其诊断老年痴呆的灵敏度与特异度。结果与正常组相比,老年痴呆患者血清HDL-C、LDL-C、Hcy、ANP水平以及MMSE评分方面均差异显著(P <0. 05),miR-9表达水平明显下降(P <0. 01),miR-34c表达水平明显上升(P <0. 01)。依据CDR评分,痴呆程度越严重,miR-9水平越低,miR-34c水平越高。老年痴呆患者血清中miR-9与LDL-C和Hcy水平均呈负相关,与HDL-C、ANP水平以及MMSE评分均呈正相关; miR-34c与血清LDL-C和Hcy水平均呈正相关,与HDL-C、ANP水平以及MMSE评分均呈负相关。ROC分析表明,miR-9和miR-34c的AUC面积分别为0. 811、0. 819;敏感度分别为89. 58%、91. 67%;特异度分别为80. 00%、71. 11%;截断值分别为0. 87、1. 14。结论老年痴呆患者血清miR-9表达下调,而miR-34c表达上调,二者异常表达均与老年呆发生及发展过程有关,可作为临床诊断和病情监测的重要指标。
Objective To investigate the expression of miR-9 and miR-34c in serum of patients with Alzheimer’s disease(AD),and to analyze their diagnostic value.Methods 73 patients with Alzheimer’s disease treated in our hospital,were selected as the subjects,at the same time,70 cases healthy examiner were selected as the normal group.The levels of serum miR-9 and miR-34c were detected by RT-qPCR in all the subjects,and the serum indexes of the two groups were compared,the sensitivity and specificity of the diagnosis of Alzheimer’s disease were observed.Results Compared with the normal group,there were significant differences in the of serum HDL-C,LDL-C,Hcy,ANP levels and MMSE score in Alzheimer’s disease(P<0.05).The level of serum miR-9 in Alzheimer’s disease patients was significantly decreased(P<0.01),and miR-34c was significantly increased(P<0.01).The serum miR-9 was negatively correlated with LDL-C and Hcy levels in patients with Alzheimer’s disease,and positively correlated with HDL-C,ANP levels and MMSE scores.The serum miR-34c was positively correlated with serum LDL-C and Hcy levels.HDL-C,ANP levels,and MMSE scores were all negatively correlated.The results of ROC analysis showed that the AUC area of miR-9 and miR-34c was 0.811 and 0.819 respectively,and the sensitivity was 89.58%and 91.67%respectively,and the specificity was 80.00%and 71.11%respectively,and the truncated values was 0.87 and 1.14 respectively.Conclusion The expression of miR-9 in the patients with Alzheimer’s disease is down-regulated,and the expression of miR-34c is up-regulated,the abnormal expression of the two is related to the occurrence and development of Alzheimer’s disease,which can be used as an important indicator of clinical diagnosis and monitoring of the disease.
