摘要
以多花黄精根茎为材料,研究外植体灭菌、不定芽分化、丛生芽增殖以及生根的最佳方法。结果表明:用0.1%HgCl2溶液处理10 min根茎组织块污染率为0;根茎组织块初代培养及不定芽增殖培养最佳培养基为MS+TDZ1.0 mg·L-1+NAA 0.5 mg·L-1+蔗糖30 g·L-1,不定芽诱导率43.3%,平均不定芽数4.62个,3次继代增殖倍数可达52.9;丛生芽生根培养前经MS+6-BA 1.0 mg·L-1+TDZ 0.5 mg·L-1+NAA 0.5 mg·L-1+蔗糖30 g·L-1增殖培养一个周期,更有利于后期生根;最佳生根培养基为1/2 MS+NAA 0.5~1.0 mg·L-1+蔗糖30 g·L-1,生根率达到77.5%,平均不定根7.36个。
The rhizome of Polygonatum cyrtonema were used to study the disinfection,adventitious bud differentiation,proliferation and rooting.The results indicated that the contamination rate of the rhizome tissue masses was0for10min treated with0.1%HgCl2solution;the optimal medium for primary culture and adventitious bud proliferation was MS+TDZ1.0mg·L^-1+NAA0.5mg·L^-1+sucrose30g·L^-1;the induction rate of adventitious buds was43.3%,the average number of adventitious buds was4.62,the triple multiple of the three generations can reach52.9;before root culture of adventitious buds,the culture was carried out for one cycle through MS+6-BA1.0mg·L^-1+TDZ0.5mg·L^-1+NAA0.5mg·L^-1+sucrose30g·L^-1,which was more conducive to the later rooting;the optimal rooting medium was1/2MS+NAA0.5-1.0mg·L^-1+sucrose30g·L^-1,the rooting rate reached77.5%,and the average adventitious root was7.36.
