Bax在普萘洛尔诱导人血管瘤内皮细胞凋亡中的作用 被引量:2

摘要 目的:通过不同浓度的普萘洛尔干预体外培养的血管瘤内皮细胞,探讨Bax基因在普萘洛尔诱导血管瘤内皮细胞凋亡中的作用。方法:体外培养人增殖期血管瘤内皮细胞,分别加入不同浓度的普萘洛尔药物干预,选择适宜的研究浓度及观察时间。构建Bax基因高表达及低表达组,检测其表达差异对于血管瘤内皮细胞凋亡的影响。结果:经普萘洛尔干预后细胞呈凋亡形态学改变,且随着药物浓度和药物作用时间的增加,细胞凋亡率也相应增加,其中300μmol/L的普萘洛尔干预24h时细胞凋亡率为28.49%,此为本实验中普萘洛尔诱导血管瘤内皮细胞凋亡的最佳药物浓度和时间点。Bax高表达组细胞构建成功后,经普萘洛尔干预,BAM7+普萘洛尔组>DMSO+普萘洛尔组>普萘洛尔组>BAM7组>空白对照组。Bax低表达组构建成功后,经普萘洛尔干预,BaxsiRNA+普萘洛尔组>N.CsiRNA+普萘洛尔组>转染试剂+普萘洛尔组>普萘洛尔组>空白对照组。结论:300μmol/L的普萘洛尔干预24h,是普萘洛尔诱导血管瘤内皮细胞凋亡的最佳药物浓度和时间点。Bax与普萘洛尔诱导血管瘤内皮细胞凋亡密切相关,Bax表达量越高,普萘洛尔诱导血管瘤内皮细胞凋亡的能力越强,Bax表达量降低,血管瘤内皮细胞凋亡无明显变化。 Objective Hemangioma endothelial cell(HemEC)which is cultured in vitro is intervened by different concentrations of propranolol.To verify the effect of Bax on propranolo induced HemEC apoptosis.Methods Different concentrations of propranolol were added to HemEC which was cultured in vitro,select the most meaningful group and time point.Adopt the Bax activator to build the high model and the low expression group.The effect of expression difference on apoptosis of endothelial cells in hemangioma was detected.Results After intervented,the HemECs performanced apoptotic morphological changes,as the drug concentration and the intervention time increased,the apoptosis rate increased too.The intervention factors that be intervented by 300μmol/L after 24 hours which apoptosis rate was 28.49%is believed to be significance.With the high expression of Bax mRNA,after propranolol intervention,BAM7+propranolol>DMSO+propranolol>propranolol>BAM7>control group.With the low expression groups,after propranolol intervention,Bax siRNA+propranolol>N.C siRNA+propranolol>transfection reagent+propranolol>propranolol>control group.Conclusion The optimal concentration and time point of pronaphthol in inducing apoptosis of endothelial cells in hemangioma was 300 mol/L of pronaphthol intervention for 24 hours.Bax gene is closely related with HemEC apoptosis induced by propranolol,the higher expression of Bax gene,the ability to induce the apoptosis is stronger.
