摘要
目的探讨通过腹腔注射鱼藤酮建立帕金森病(Parkinson’s disease,PD)大鼠模型,观察其行为学与病理损伤的改变。方法雄性Wistar大鼠随机分为正常对照组、生理盐水对照组、溶剂注射组和鱼藤酮模型组。生理盐水对照组、溶剂注射组与鱼藤酮模型组大鼠,连续21天分别腹腔注射生理盐水、溶剂和鱼藤酮。监测各组大鼠体重的变化,比较各组大鼠行为学(方桥实验、旷场实验)的变化,观察中脑黑质酪氨酸羟化酶(tyrosine hydroxylase,TH)免疫阳性神经元数量与TH蛋白表达水平的改变。结果与各对照组相比,鱼藤酮模型组大鼠体重显著性下降,方桥存留时间和运动速度显著性降低,中脑黑质TH(+)神经元数目和TH蛋白表达水平也显著性降低。结论采用鱼藤酮腹腔注射的方法可以成功制作PD大鼠模型,该模型具有明确的PD类似的行为学和病理损伤,为PD进一步的研究提供了新的途径。
Objective: To make the model of Parkinson’s disease( PD) rats by rotenone intraperitoneal injection,and observe its changes in behavioral test and pathology. Methods: Male Wistar rats were randomly divided into four groups on average( normal control group,saline control group,vehicle control group and rotenone model group). Rats of saline control group( SCG),vehicle control group( VCG) and rotenone model group( RMG) were administered with saline,vehicle and rotenone through daily intraperitoneal injection for 21 days,respectively. Changes of weight and behavior,including square bridge test and open field test were measured in different groups. Changes of the number of tyrosine hydroxylase-immunoreactive( TH-ir) neurons and expression of TH in substantia nigra( SN) were also measured in different groups. Results: The weights in rats of RMG were decreased. The fallen latency in square bridge test and the average moving speed in open field test were reduced in rats of RMG. The number of TH-ir neurons and expression of TH in SN of RMG rats were also reduced. Conclusion: Model of PD rat by rotenone intraperitoneal injection is successfully established. It has similar motor and pathologic deficits with PD. This model may provide a new pathway to further related studies of PD.
