动物源性食品检测磺胺类残留ELISA试剂盒的研制

Development of ELISA kit for detection of sulfonamides residues in animal derived foods

为开发一种能够稳定快速检测磺胺类药物残留,以自制抗原和单克隆抗体为材料,建立一种ELISA试剂盒快速检测方法。结果表明,抗原包被浓度2×104倍,抗体∶酶标二抗为2×104∶2000;方法灵敏度为3. 88 ng/m L,检测范围1. 18～41. 67 ng/m L,可以检测大多数磺胺类似药物,4℃试剂盒保存180 d参数稳定。样品进行的添加回收实验,磺胺类药物及其磺胺类似物回收率在67. 4%～117%,批间变异系数<10%,批内变异系数<10%。

In order to develop a stable and rapid detection method for sulfonamides residues,a rapid detection method of ELISA kit was established by using self-made antigens and monoclonal antibodies. The results showed that the antigen concentration was 2 × 104 times,and the antibody to enzyme standard two was 2 × 104: 2000; the sensitivity of the method was 3. 88 ng/m L,Detection range 1. 18 ng/m L-41. 67 ng/m L,which could be used to detect most sulfonamides like drugs,and the kit 180 d parameters were stable at 4 ℃. The recoveries of sulfonamides and their sulfonamides were 67. 4%-117%,and the coefficient of variation among batches was < 10%,and the coefficient of variation within batch was < 10%.
